Objective:To study the effect and mechanism of hypericin on human lung cancer cell line H460.Methods:H460 cells were cultured in vitro,and the high,medium and low concentration drug serum group and positive drug(cisplatin)containing serum group,to detect the serum MTT value method and effect on human H460 cells;in the serum were detected by flow cytometry technique under the condition of 460 lung cancer cell apoptosis effect;Western Blot method for detection of human lung cancer cells in H460 Active Caspase-3,bcl-2,Bax protein showed the change.Result:1 MTT results showed that hypericum attenuatum group serum compared to blank group could significantly inhibit human lung cancer cell line H460 proliferation,and with the time(72h)and the concentration of drug serum increased the inhibition gradually increased.In the three groups of high,medium and low levels,the highest effect of the high dose group was found.2 flow cytometry showed that the cell apoptosis,H.attenuatum chosiy group compared to the blank group containing serum could significantly promote the apoptosis of human lung cancer cell line H460;H.attenuatum chosiy high,medium and low dose group compared to three,H.attenuatum chosiy high dose group to promote apoptosis of H460 cells to the most significant.3 the results of Western Blot test showed that the serum levels of Active,Bax,protein and down regulated expression of bcl-2 protein were significantly increased in the serum of each group compared with the blank group.Conclusion:1 the serum contained in the high,medium and low serum levels of all of them could inhibit the proliferation and promote the apoptosis of H460 lung cancer cells.2 the mechanism of apoptosis in human lung cancer cell line H460 induced by the serum contained in each group was related to the death receptor and mitochondrial signal transduction pathway.
