| Background:To study the effect of Ding’s kuijie enema by analyzing its implication for the expression of Occludin,MUC2mRNA,iNOSmRNA in colonic tissue and serum IgA of rats with ulcerative colitis induced by DSS,and to explore its possible Mechanism.Methods:Ten SD rats were randomly selected as normal control group,and the remaining 50 SD rats were established by DSS.After the establishment of the model,the rats were randomly divided into 5 groups:model group,Ding’s kuijie enema low dose group,Ding’s kuijie enema middle dose group,Ding’s kuijie enema high dose group and Mesalazine enema group.In addition to the normal control group,the other groups were given saline and the corresponding drugs enema for 14d.After 14 days of treatment,the rats were sacrificed by the method of taking the abdominal aorta and the colon specimens were taken,observed and scored the changes of body weight,stools,blood in the stools,evaluate the disease activity index,the general situation and pathological changes of colonic tissue were observed and scored respectively in visual observation and under light microscope.The levels of serum IgA were measured by ELISA,and the Occludin protein content in colonic tissue was detected by Western Blot.Real-time PCR was used to detect the expression of MUC2 and iNOS mRNA in colon tissues.Results:1.After modeling,rats are apathetic,weight loss,stool sugar with mucus pus and blood,compared with the normal control group,the DAI score increased(P<0.05).After the enema treatment,the above symptoms were improved,compared with the model group the DAI score decreased(P<0.05),the low dose group were less than those in the other groups,but better than the model group.2.Visual observe the rats in the model group showed incomplete injury,obvious congestion and edema,and associated with ulcer formation,Compared with the normal control group,the score was increased(P<0.05).The rats treated with Ding’s kuijie enema and Mesalazine enema congestion,edema and mild erosion,and no obvious ulcer formation was observed in the colonic mucosal,and the scores were significantly lower than those in the model group(P<0.05).3.Under the light microscope,the rats in the model group had necrotic shedding after colonic mucosal injury,accompanied by varying degrees of ulcer foci,infiltration of inflammatory cells,mucosal glands were destroyed,arranged in sparse disorder,or even absent,compared with the normal control group,the pathological score was significantly increased(P<0.01).There were no significant ulcer formation,mucosal edema and inflammatory cell infiltration in the colonic mucosa of rats in Ding’s kuijie enema group and Mesalazine enema group,compared with the model group,the pathological scores were significantly decreased(P<0.01).4.The level of serum IgA and the expression of Occludin protein in the model group were significantly lower than those in the normal control group(P<0.05).After treatment of Ding’s kuijie enema and Mesalazine enema,the level of serum IgA and the expression of Occludin protein were significantly higher than those in the model group(P<0.05),.5.The expression level of iNOS mRNA in colon tissue of model group was significantly higher than that of normal control group(P<0.01).After treatment of Ding’s kuijie enema and Mesalazine enema,the expression of iNOS mRNA were decreased compared with that of model group(P<0.05).The expression level of MUC2 mRNA in colonic tissue of model group was significantly lower than that of blank control group(P<0.01),after treatment of Ding’s kuijie enema and Mesalazine enema,the expression level of MUC2 mRNA was increased compared with that of model group(P<0.05).There was no significant difference among middle high dose group and Mesalazine enema group in the comparison of mRNA expression level.Conclusions:1.Ding’s kuijie enema have a certain therapeutic effect on DSS-induced UC,can effectively reduce its stools,blood in the stool,weight loss and other symptoms,improved intestinal mucosal inflammation,reduce the DAI score and general and pathological score.2.The intestinal injury of DSS-induced UC,Ding’s kuijie enema can significantly increase Occludin protein,MUC2 mRNA expression levels,serum IgA content,reduce iNOS mRNA expression levels.Reduce intestinal mucosal inflammation in rats from the intestinal mechanical barrier,immune barrier and chemical barrier. |
