Peripheral nerve injury is a common traumatic disease in clinic.Despite the rapid development of microsurgery,the functional recovery after nerve repair has been improved,but the actual effect is still difficult to reach the ideal level.In order to better and faster promote the recovery and regeneration of nerve,many scholars have carried out a lot of basic research and clinical application.At present,more clinical use of Western medicine based,these commonly used drugs have limited efficacy,and expensive,heavy economic burden.Therefore,to give full play to the advantages of traditional Chinese medicine and the combination of traditional Chinese and Western medicine,it is necessary to explore the highly effective and low toxicity drugs for the repair of nerve injuries and elucidate their molecular mechanisms,which is of positive significance for clinical treatment.The molecular mechanism of this experiment is to study the leech effect on damaged peripheral nerve repair,especially on nerve growth factor and brain-derived neurotrophic factors,nerve growth associated protein related gene expression of qualitative and quantitative research.ObjectiveThe simulation model of rat sciatic nerve injury treated by intragastric administration of Hirudo preparations,observation and nerve recovery in rats with growth factor brain-derived neurotrophic factor,nerve growth associated protein expression of related gene.MethodsSelected 100 SD rats,the preparation of the left sciatic nerve injury model rats were randomly divided into 4 groups,namely leech group,Methycobal group,saline control group and blank control group.Leech group,Methycobal group,saline group for prevention of postoperative infection in 3 days,fourth days were given water decoction of leech and Mecobalamin tablets dissolved in water and saline solution.After fourth,eighth weeks,to observe the nerve tissue injury recovery under electron microscope,and take the cell apoptosis detected by TUNEL L5 in dorsal root ganglion,protein expression and immunohistochemistry to detect NGF,BDNF,GAP-43.ResultsThe recovery of sciatic nerve tissue in leech group was better,and the expression of NGF,BDNF and GAP-43 were better than that of control group,but the number of apoptotic cells was less than that of control group,and the difference was statistically significant.ConclusionsLeech may be through the promotion of NGF and BDNF and GAP-43 repair related nerve growth factor expression by growth repair,inhibition of apoptosis,play a protective effect on sciatic nerve injury in rats.
