| Objective:Discuss sevoflurane function in rats after femoral artery thrombosis have influence and its mechanism of action.Methods:Selects the weight for 250-300g,fourty SD rats(♀=25,♂=25)were randomly divided into control group(C group),model group(M group),sevoflurane low concentration group(SL group)and high concentra thrombosis model group(SH group).The control group except,the rest of the three groups were established by using ferric trichloride artery thrombosis model.The different concentra of sevoflurane(3%,and 4%)were continuous infused two hours via caudal vein in the SL and SH groups,the control and model groups were continuous suction air two hours.The weight of common femoral arterial thrombus were measured,observe the pathologic changes of the femoral artery of rats by HE staining.The PF4、vWF、AT-Ⅲ、P-selsct、BCL-2 and BAX of plasma and liver was examined by Elisa assay.The protein levels of BCL-2 and BAX in femoral artery was detected by Western blot method.Results:(1)The results of the thrombus weight weighed:and the model group to compared,the weight of common carotid arterial thrombus were significantly decreased in the different concentration of sevoflurane groups(P<0.05 or P<0.01).(2)HE staining results indicated that there was no obvious pathological changes in the control group,the arterial lumen was completely blocked by thrombus and vascular endothelial cells was seriously stripped in the model group,the low concentra of sevoflurane group,the arterial lumen was more blocked,endothelial cells was heavily stripped.The high concentra of sevoflurane group,the arterial lumen was less blocked,endothelial cells was slightly stripped.(3)ELISA:[1]The detection results of the liver:1.And the control group to compared,the levels of PF4 and vWF were significantly increased(All P<0.05);And the model group to compared,the levels of PF4 and VWF were content decreased obviously in the different concentra of sevoflurane groups(P<0.05).2.And the control group to compared,the levels of P-selectin and AT-Ⅲwere significantly increased(All P<0.05);And the model group to compared,the levels of P-selectin and AT-Ⅲwere significantly decreased(P<0.05,P<0.01),in hing concentration of sevoflurane groups the levels of P-selectin were significantly decreased(P<0.01).3.And the control group to compared,the model group BAX and BCL-2 significantly increased(P<0.01);And the control group to compared,the levels of BCL-2 significantly decreased(P<0.05)in the different concentra of sevoflurane groups,but the levels of BAX significantly decreased(P<0.01).[2]The detection results of the plasma:1.And the control group to compared,the levels of PF4 and vWF were significantly increased.Compared with the model group,the levels of PF4 and VWF were content decreased obviously in the different concentra of sevoflurane groups(P<0.05).2.And the control group to compared,the model group the levels of BAX and BCL-2 were significantly increased(P<0.01);And the model group to compared,the levels of BAX and BCL-2 were significantly decreased in the different concentra of sevoflurane groups(P<0.01).[3]The changes of the expression of BCL-2and BAX protein:And the control group to compared,the expression of BCL-2and BAX protein were significantly increased in the model group;And the model group to compared,the expression of BCL-2/BAX significantly decreased.The changes of the expression of BCL-2and BAX protein were significantly decreased in the different concentra of sevoflurane groups(All P<0.01).Conclusion:(1)Sevoflurane has inhibitory effect on the formation of the femoral artery.(2)The sevoflurane inhibit thrombosis may be by protecting vascular endothelial cells,the mechanism of preventing platelet aggregation,,and inhibiting apoptosis. |
