Purpose:To prepare a kind of drug delivery system loading docetaxel with absorbable,degradable,inexcitant and controlled released natural polymer materials Bletilla striata gels and study their antitumor effects via Intraperitoneal injection.Methods:1,Take suitable amount of dried Bletilla striata.Then extracted and purified Bletilla striata.The content determination of the docetaxel-loaded Bletilla striata gels was measured by anthrone-sulfuric acid method.Then Bletilla striata polysaccharide was dissolved by pured water.Prepare the docetaxel-loaded Bletilla striata gels via Physical methods.The characterations of docetaxel-loaded Bletilla striata gels were evaluated by scanning electron microscope and carried on the in vitro releasing experiments.2,H22 cells were intraperitoneally injected into ICR mice to develop the ascites tumor models.After 24h when the models developed,15 mices were randomly divided into three groups:normal saline contrast,docetaxel,docetaxel-loaded Bletilla striata gels.The docetaxel and docetaxel-loaded Bletilla striata gels groups were treated i.p.with 10mg/kg of docetaxel.The normal saline contrast group was given i.p.with same volume saline.All the treatments were given 4 times and the interval was 72 hours.In the period,the general state of mice were observed.All mice were killed on the 15th day after the treatment.Collected all the ascites,evaluate the amount of ascites and observed the peritoneal metastasis nodules.Important organs of docetaxel-loaded Bletilla striata gels group were taken.Sliced the embeded paraffin to observe each viscera injury after HE staining.Results:1,The content determination of the docetaxel-loaded Bletilla striata gels was 67.26%.Docetaxel was observed average distributed in Bletilla striata gels by scanning electron microscope.In vitro releasing experiments showed 45%of the docetaxel-loaded Bletilla striata gels were released after 150 hours.2,10 days after the treatment,measuring weights of all groups,the normal saline contrast group was much heavier than other two groups(P<0.05);The amount of ascites was less in the docetaxel-loaded Bletilla striata gels group than the normal saline contrast group(P<0.05).There were much less peritoneal metastasis nodules in the docetaxel-loaded Bletilla striata gels group than the others.Important organs of docetaxel-loaded Bletilla striata gels group were not injuried observed by the electron microscope.Conclusion:The preparation of docetaxel-loaded Bletilla striata gels were simple.Docetaxel was averagely distributed in Bletilla striata gels.The docetaxel-loaded Bletilla striata gels were slowly released.Intraperitoneal docetaxel-loaded Bletilla striata gels showed certain anti-tumor effect.The docetaxel-loaded Blenlla strziata gels deserves further research. |
