Study On The Correlation Between Over-expression Of Heparanase Gene And Syndecan-1 Expression In Gallbladder Carcinoma Cell Lines In Vivo | Posted on:2018-03-12 | Degree:Master | Type:Thesis | Country:China | Candidate:H Jin | Full Text:PDF | GTID:2334330515456213 | Subject:Surgery | Abstract/Summary: | PDF Full Text Request | Objective:To discuss the correlation between over-expressed heparanase gene and syndecan-1 expression in gallbladder carcinoma cells in vivo in rat models and also the different invasion ability of normal gallbladder carcinoma cells and heparanase gene over-expressed gallbladder carcinoma cells in vivo in rat models.Method:1.To observe the growth of normal gallbladder carcinoma cells and heparanase gene over-expressed gallbladder carcinoma cells in vivo in rat models.2.Western-blot methods are applied to detect the protein expression of Syndecan-1 in GBD-SD and GBC-SD.3.RT-PCR technologies are applied to detect the Syndecan-1 gene expression quantities in GBD-SD and GBC-SD.4.Immunohistochemistry methods are applied to detect the protein expression of Syndecan-1 in GBD-SD and GBC-SD.Result:1.GBD-SD and GBC-SD gallbladder carcinoma cells grows in character in vivo in rat models.2.The expression of Syndecan-1 protein in GBD-SD was decreased sharply compared to Syndecan-1 in GBC-SD,and the difference was statistically significant(P<0.05).3.The m RNA of Syndecan-1 in GBD-SD significantly reduced than that of GBC-SD,the difference was tatistically significant(P<0.05).4.The protein amount of Syndecan-1 in GBD-SD were decreased sharply compared to protein amount of Syndecan-1 in GBC-SD,and the difference was statistically significant(P<0.01).Conclusion:1.In gallbladder carcinoma cells in vivo,a negative correlation was detected between the expression of Heparanase and the amount of Syndecan-1.And a positive correlation was detected between the invasion ability of gallbladder carcinoma cells and theexpresion of heparanase.2.In gallbladder carcinoma cells in vivo,as the expression of Heparanase was increased,the expression of Syndecan-1 was decreased,and the invasion ability was enhanced.The Heparanase/Syndecan-1 axis played an important role in the invasion and infiltration of gallbladder carcinoma.3.Heparanase/Syndecan-1 axis is going to be applied to the diagnosis and treatment of gallbladder cancer.At the same time,the axis of Heparanase/Syndecan-1 is promising to provide new theoretical and experimental basis for anti-cancer drugs.However,the axis of Heparanase/ Syndecan-1 of gallbladder carcinoma still needs further research. | Keywords/Search Tags: | Heparanase, over-expression, Syndecan-1, gallbladder carcinoma, correlation, vivo | PDF Full Text Request | Related items |
