| Objective:To observe the effect of the blood lipid in the precautionary high-fat model Wistar rats were given by gavage Agriophyllum,Agriophyllum aqueous extraction and Agriophyllum alcohol extraction.Filter the active ingredients of the hypolipidemic and determination of content and determination of molecular weight of the Agriophyllum polysaccharide.Methods:1.Using precautionary high-fat model,measure TC,TG,LDL-C,HDL-C concentration by automatic biochemical instrument after gavage Agriophyllum,Agriophyllum aqueous extraction and Agriophyllum alcohol extraction 15 days later.2.Using precautionary high fat model,measure TC,TG,LDL-C,HDL-C concentration by automatic biochemical instrument after gavage Agriophyllum water-soluble small molecule and Agriophyllum crude polysaccharide 15 days later.The liver of the rats were examined in pathology.Through an acute toxicity experiment,the safety of the small molecules of water soluble in the Agriophyllum and the crude polysaccharides of Agriophyllum was preliminarily studied.3.Use UV spectrophotometer to measure the total polysaccharide content of the Agriophyllum.The molecular weight of the polysaccharide was determined by gel chromatographyResults:1.Compared with the high-tat model group,the serum TC concentration(P<0.01)could be significantly decreased in the positive control group,the water extraction group and the alcohol extraction group.The concentration of serum TC of Agriophyllum original drug group were decreased(P<0.05).Compared with the high-fat model group,the serum TG concentration(P<0.01)could be significantly decreased in the positive control group,the original drug group,the water extraction group and the alcohol extraction group.The LDL-C concentration was significantly lowered in the positive control group,the alcohol extraction group and decreased in the original drug group and water extraction group(P<0.05).Compared with the high-fat model group,there was a significant increase in HDL-C concentration,but there was no significant difference between the positive control group,the original drug group,the water extraction group and the alcohol extraction group.2.TC,TG and LDL-C in rats of crude polysaccharide administration group were significantly lower than those in the model control group(P<0.01).There was no significant difference in high serum lipoprotein cholesterol(HDL-C)between the Agriophyllum small molecule and polysaccharide groups.Liver pathological observation of rats showed that:polysaccharides can make hyperlipidemia in rats to reduce the degree of fat deposition in the liver,help to prevent hyperlipidemia.The rats did not die in 216 times the daily dose of the drug,and the result showed that the water-soluble small molecules and the crude polysaccharide were low in toxicity.3.Phenol-sulphate spectrophotometry to determine the total polysaccharides content of the Agriophyllum.The average content of polysaccharides was 113.15mg·g-1(n=6).Determined by high performance gel chromatography(GPC),the number average molecular weight of polysaccharide was 58771 and the weight average molecular weight was 66489.Conclusuion:Through animal experiments,the lipid effect of the Agriophyllum was found.It has the effect of lowering total cholesterol,triglycerides,and low density lipoprotein.Through the filter of the active ingredient,the composition of the lipid was found to be the crude polysaccharide,and the content and the molecular weight were studied. |
