Objective: To study the effection of RORα/γ inverse agonist on Th1,Th2 and Th17 cells in a murine model of allergic rhinitis and further explore the possibility to cure allergic rhinitis by inhibiting of RORα/γ transcription factors.Methods: Eighteen-six week-female C57BL/c mice were randomly divide into three groups: control group,AR group and SR1001 group.After the establishment of mouse models,the total emotions of sneezing and rubbing were counted to evaluate the nasal symptoms among groups;H&E and PAS staining were used to observe the Eosinophils and goblet cells in nasal tissues;the percentage of Th1,Th2,and Th17 cells in spleen were measured by flow cytometry;the IFN-γ,IL-17 and OVA-sIgE in serum and the IL-5,IL-13 in nasal fluid lavage were measured by ELISA;immunohistochemical technique was adopted to measure the changes of ST2 positive cells in nasal mucosa.Results: 1.The total emotions of sneezing and rubbing in AR group were significantly higher than the control group,while the emotions were lower in SR1001 group than the AR group(P<0.05).2.The percentage of Th2 and Th17 cells in spleen,IL-5,IL-13 levels in nasal fluid lavage and IL-17 levels in serum in AR group were significantly higher than the control group,and lower in SR1001 group compared with the AR group(P<0.05).3.The percentage of Th1 cells and Tbet in spleen,the IFN-γlevels in serum were significantly lower in AR group than control group,and higher in SR1001 group compared with AR group(P<0.05).4.The ST2 positive cells were significantly higher in AR group than control group,and lower in SR1001 group compared with AR group(P<0.05).Conclusions: 1.RORc inverse agonist can alleviate the nasal symptoms of sneezing and rubbing in AR mice.2.RORc inverse agonist can suppress Th17 response in AR mice.3.RORc inverse agonist can promote Tbet associated Th1 responses but inhibit Th2 responses in AR mice.4.RORc inverse agonist can suppress the expression of ST2 in nasal mucosal thus inhibit a ILC2 associated responses.
