Background: Diabetes mellitus is a metabolic integrated disease in the body with chronic glucose imbalance at the glucose concentration level that is characterized by normal blood glucose diseases that endanger human health.So far,there is no way to completely cure it.With the application of new technologies such as genomics and proteomics,it provides a new way to relieve and treat diabetes mellitus.According to the existing study found that NNMT in adipose tissue and liver tissue is highly expressed,and NNMT overexpression can damage the β cells caused by abnormal insulin secretion,which may be an important molecular mechanism of diabetes,and exercise can be reduced Weight and insulin resistance,regulate lipid metabolism,study the effect of different exercise patterns on NNMT expression in type 2 diabetic rats,and to further understand the effect of exercise on the pathogenesis of diabetes mellitus,so as to provide more scientific and effective treatment for the treatment of type 2 diabetes mellitus Exercise therapy.Objective: To observe the effects of different exercise patterns on the indexes of body weight and blood glucose in rats with type Ⅱ diabetes mellitus.To observe the expression of NNMT in different exercise pattern rats,and to provide scientific basis for exercise therapy of type Ⅱ diabetes mellitus.Methods: 44 SD healthy male rats were randomly selected 8 rats as healthy control group(HC group),fed with basic diet,and the remaining 36 were fed with the basal diet for one week And then fed with STZ(streptozotocin)in rats.The rats were randomly divided into three groups.The rats were randomly divided into three groups:DC group),diabetic aerobic exercise group(DOC group),diabetic anaerobic intermittent exercise group(DAI group),each group of 8,each group were fed with basic feed.Then the exercise group was subjected to 6 weeks of different exercise mode intervention,aerobic continuous exercise group treadmill speed of 15 m / min,anaerobic intermittent exercise group running speed of 25 m / min.The training frequency is 6 days per week and the exercise time is 40 minutes.Weekly trainingafter fasting weighing to take tail vein blood fasting blood glucose.After 6 weeks of training all the end,with ether anesthesia,with EDTA tube to take the right atrial venous blood 1.5-2ML,room temperature for 30 min after coagulation 2000 r centrifugation 15 min,separation of serum,marked and put in-80 ℃ refrigerator to be kept The And then immediately take the liver,fat,and pancreas and other organizations,wrapped with tin foil,do a good job,and then placed in a container filled with liquid nitrogen,and then into the-80 ℃ refrigerator to be tested.Serum insulin and glycated hemoglobin were measured by ELISA kit.The content of NNMT in each tissue was detected by Western Blot immunoblotting.Result:(1)After 6 weeks of training,the blood glucose of DAI group was significantly lower than that of DC group and DOC group(P <0.01),and it was also significantly decreased compared with that before exercise(P <0.01).After 6 weeks of training,the glycosylated hemoglobin of DAI group was also significantly decreased compared with DC group and DOC group(P <0.05).(2)After 6 weeks of training,the insulin sensitivity index of DAI group was significantly higher than that of DC group and DOC group(P <0.05).Although DOC group and DC group also increased,the difference was not significant(P <0.05).Although there was a decrease in the insulin resistance index between the D group and the DC group and the DOC group(P <0.05),the difference between the DOC group and the DC group was not significant.(3)The expression of NNMT in the liver of all diabetic rats(DC,DOC and DAI)was significantly higher than that in HC group(P <0.05).After 6 weeks of training,the expression of NNMT in DAI group was significantly lower than that in DC group(P < 0.05).The expression of NNMT in the liver of the DOC group was not significantly different from that in the DC group(P> 0.05).The expression of NNMT in the muscle tissue of the exercise group was significantly higher than that in the DC group(P <0.05)DAI group was slightly higher than DOC group,but the difference was not significant(P> 0.05).Conclusion:(1)anaerobic intermittent exercise with significant hypoglycemic and hypoglycemic hemoglobin effect,and aerobic exercise hypoglycemic and hypoglycemic hemoglobin effect is not obvious.(2)Anaerobic intermittent exercise has obvious effect on improving the insulin sensitivity of diabetic rats and alleviating insulin resistance in diabetic rats,and the effect of aerobic exercise is not obvious.(3)The level of NNMT protein expression in the liver was almost identical with that offasting blood glucose and insulin resistance index.The significant inhibitory effect on NNMT protein expression in the liver may be the main reason for anaerobic intermittent transport to more effectively reduce blood glucose and alleviate insulin resistance.
