| Objective: To study the learning and memory ability and the expression of ERK1 and Caveolin-1 proteins in the hippocampus of rats with kidney deficiency,and to investigate the molecular mechanism of the decline of learning and memory in the kidney deficiency from the point of view of hippocampal synaptic plasticity.Methods: 20 SD rats were randomly divided into two groups: blank group(4 males and females each)and kidney deficiency model group(6 males and females each).In the cage mating mode,the "cat scare mice" model was used to frighten the pregnant rats of the kidney deficiency model group until the offspring were produced.After 4 weeks,the blank group randomly selected 12 rats as blank group,a total of 48 pups were randomly selected from the kidney deficiency model group as model group,Bushen group,Shugan group,heart group.12 rats in each group.Intimidate the model group daily,intimidation of Bushen group,Buxin group,Shugan group and drug intervention.Bushen group with Six Ingredient Rehmannia Pill liquid,Buxin group with Tianwangbuxin Dan medicine liquid,Shugan group with Xiaoyao Pill liquid,for 3 consecutive months.The body weight and brain weight of each group were measured.The expressions of ERK1 and Caveolin-1 proteins in hippocampus were measured by immunohistoch emistry.Results:(1)Morris water maze navigation experiment:compared with the blank group,model group’s escape latency was prolonged(P <0.01);compared with the model group,escape latency of Bushen group,Buxin group decreased sign ificantly(P <0.01),Shugan group did not decrease significantly,no statis tical significance;compared with Buxin group,Bushen group escape latency was shortened(P <0.05);compared with the Shugan group,Bushen group escape latency decreased significantly(P <0.01).Compared with the blank group,model group total distance increased significantly(P <0.01);compared with the model group,total distance of Bushen group,Bushen group group,Shugan group were significantly reduced(P <0.01);compared with Buxin group、Shugan group,Bushen group’s total distance decreased significantly(P <0.01).Space exploration experiment,compared with the blank group,the rats in the model group 30 s for the first time through the target area was prolonged(P <0.01);compared with the model group,Bushen group,Bushen group,Shugan group for the first time through the target area time was shortened(P <0.01);compared with Buxin group,Shugan group,Bushen group’s first across the target area time was shortened(P <0.05).Compared with the blank group,the rats in the model group 30 s within the target area of residence time was significantly reduced(P <0.01);compared with the model group,target area residence time of Bushen group,Buxin group,Shugan group increased significantly(P <0.01);compared with Buxin group,Shugan group,Bushen group’s target area of residence time obviously extended(P <0.01).(2)The body weight and brain weight of rats: compared with blank group,the weight of the model group significantly reduced(P <0.01);compared with the model group,weight of Bushen group,Bushen group,Shugan group increased significantly(P <0.01);compared with Buxin group,Shugan group,Bushen group’s body weight increased significantly(P <0.01).Compared with the blank group,model group brain weight reduced significantly(P <0.01);compared with the model group,brain weight of Bushen group,Bushen group,Shugan group increased significantly(P <0.01);compared with Buxin gro up,Shugan group,Bushen group’s brain weight increased(P <0.05).(3)The frozen section immunohistochemistry:compared with the blank group,model group’s hippocampal ERK1/ Caveolin-1 immunoreactive cell number was significantly decreased(P <0.01);compared with the model group,the number of positive cells of Bushen group,Buxin group,Shugan group were significantly increased(P <0.01);compared with Buxin group,Shugan gro up,Bushen group’s number of positive cells increased significantly(P <0.01).Conclusion:The compound kidney deficiency animal model was established by the method of "congenital deficiency + acquired danger",to a certain extent reflects the formation mechanism of kidney deficiency,kidney deficiency decreased the feasibility of operation;quality of learning memory ability of rats,the mechanism might be related with decreased expression of synaptic plasticity related proteins ERK1,Caveolin-1;group learning the memory ability were improved,and tonifying kidney group was improved significantly, and the prescription may significantly improve the expression of hippocampus ERK1,Caveolin-1 protein;Buxin Decoction and Shugan prescription improvement inferior to Bushen Decoction,kidney and highlights the relationship between learning and memory,consistent with the theory of the kidney Storing Aspiration. |
