Influence Of Danzhi Xinoyao Powder On TGF-α、EGFR In Polycentric Ovary Hypeiandrogenism Rats

ObjectiveTo observe the effect of Danzhi Xinoyao Powder（DZXYP）on the expression of TGF-a Rand EGFR in rats with polycentric ovary（PCO）model built by DHEA and brief discuss its function mechanism.MethodsWe selected sixty SD female rats of twenty-one-days old who should have the regular estrum of SPF standard and randomly divided into blank group and model group.Subcutaneous inject DHEA on model group rats posterior neck to establish PCO model and subcutaneous inject soybean oil on blank group rats.We total need 21days to complete this step.After the succeed decision of molding,the model group rats were randomly divided into 3 groups:DZXYP-treated group,Diane-treated group,model control group,each group have 10 rats.DZXYP-soup and Diane-soup were respectively given by gavage in DZXYP-treated group and Diane-treated group rats.The blank control group and the model control group were given a gavage of distilled water of the same volume with the drug.We calculating the gastric volume according to the weight of each group rats every day.The lavage were continuous for21 days,seven days off,a period of treatment is 28 days,for a total of three course of treatment.Ovaries were harvested from the rats for histomorphological observation.The levels of T,LH,FSH were determined by radio-immunity.The expression of TGF-a R and EGFR were tested by immunohistochemical method.Results（1）PCO rat model assessment:After constructing the PCO model,the body weight in the model group were obviously increased compared with the normal group（P<0.01）.Compared with the normal group,the serum of T in the model group were increased（P<0.01）,moreover the serum of E₂ were decreased（P<0.01）.Most model rats oestrous cycle were in estrogen interphase which means with anovulation.Model rats ovary presented sack-like structure and follicles-expansive change under light microscope.（2）The serum of sex hormone changes（T,FSH,LH）in PCO rats after drug intervention:Compared with the normal group,the serum of T in the model group were increased significantly（P<0.01）.Compared with the model group,the serum of T in the Danzhi Xiaoyao Powder group and Diane-35 group were obviously decreased（P<0.01）.There was no significant difference in 4 group with the serum of FSH and LH（P>0.05）.（3）The local ovarian granulose cells expression of TGF-a R and EGFR protein changes after drug intervention:Compared with the normal group,the local ovarian granulose cells expression of TGF-a R and TGF-a R protein in the model group were significant decreased（P<0.01）.The local ovarian granulose cells expression of TGF-a R and EGFR protein in the Danzhi Xiaoyao Powder group and Diane-35 group were increased compared with the model group after drug intervention（P<0.01）.（4）The morphological changes in ovarian tissue of rats after drug intervention:We can see antral follicle and graaffian follicle in the normal group rats ovaries,some ovulation can be also visibled.There were many small follicle,atretic follicle and follicular dilation of follicle in the model group rats ovaries,but anovulation.The atretic follicle and follicular dilation of follicle in the Danzhi Xiaoyao Powder group and Diane-35 group rats ovaries were decreased compared with the model group after drug intervention.Some ovulation can be visibled in the Danzhi Xiaoyao Powder group.ConclusionDZXYP can reduce the levels of T and decrease the expression of TGF-a R,EGFR in rats with PCO.And its can also improve the ovarian function and promote the normal growth and development of follicle to overall improve ovary polycystic changing and ovulation barrier.It is suggested that DZXYP treat PCOS may be closely associated with the reduction of androgen and the expression of TGF-a R、EGFR in ovarian granule cell.
