Cancer stem cell(CSC)hypothesized that cancer was caused by a small subpopulation of cancer stem cells which were original drive of tumor abnormal proliferation,invasion,metastasis,drug resistance and recurrence.In this investigation,hub genes related of the cancer stem cells were excavated by analyzing the mixed expression data of genes and miRNAs in four types cancerous.Firstly,expressions profile data of 18409 genes and 1035 miRNA across 746 breast cancers are clustered into different modules by the bioinformatics methods.Furthermore,gene sets enrichment analysis in MsigDB shows the involvement of embryonic stem cell(ESC)properties with a gene set of 2019.On the other hand,a gene set of 859 genes is enriched with mesenchymal stem cell(MSC)features.In addition,the other three cancers(Glioblastoma Multiforme,Lung Adenocarcinoma and Brain Lower Grade Glioma)are used as the comparison data in order to optimized two gene sets.Finally,178 common ESC genes and 7 common MSC genes been observed in those four type cancer data.In order to verify the accuracy of two gene sets,we investigate the expression pattern of two gene sets in the merged gene expression profiles data of GSE29625 and GSE28974.Moreover,178 common ESC genes are involved of E2 F mediated transcription programsIn the end,their gene regulatory networks show that two new hub genes TPX2 and MCM10 which associated with ESC and a key regulatory gene COL5A2 which associated with MSC.Furthermore,examining the out-degrees of 178 duplicate genes correlated of embryonic stem cells in four types of cancer regulatory networks,two key regulatory genes BUB1 B and NCAPH are screened out.These genes might be used as the potential therapeutic targets for the treatment of cancer.
