Seasonal influenza often occur in Spring and winter.It brings serious influence on human body in all over the world.Currently two strains of avian influenza virus have mutated that could infect human,and lead them to death.Nowadays,ordinary vaccination and antiviral drugs would not be the only two ways to prevent influenza.That’s why broadly influenza neutralizing antibody provides a new treatment method that has obtained a great achievement.The mentioned progress has been developed in some parts,such as the development of broadly influenza vaccine,the basic research of the discovery of new common conservative sites,and so on.In the following hybridoma technique,which makes some influenza monoclonal antibodies,will be considered based on some experiments.These experiments aimed to identify the basic qualities of these antibodies.1 The preparation of influenza monoclonal antibodies.influenza A viruses such as H1N1 and H3N2,H9N2 and influenza B vir uses were used to immunize mice respectively.Each virus were used to immu-nize three mice.After three times,hemagglutination inhibition test(HAI)was utilized to assay the hemagglutination inhibition titer of serum antibodies for each virus.When those titers are above 40,cell fusion could be processed.Three days before fusion,those mice should be immunized again to activate the memory cells.Converted memory cells into activated B cells which coul d secrete antibodies.After four or three days,That hybridomas could be clon e-d is from the positive holes,which are preliminarily screened by holehemagg 1-utination inhibition.It could be collected that antibodies are produced by a single-cloned hybridoma.Fin-ally monoclonal antibodies,such as 20C5,20H7,5F7,3F7,9E6,19C9,4G4,4E4,10B6,2F7,are selected by hemagglutination inhibition.These antibodies have potential hemagglutination inhibition activity influenza virus.2 The preliminary identification of influenza monoclonal antibodies.he preliminary identification of those monoclonal antibodies could be ana-lyzed by micro-neutralization and hem-agglutination inhibition.Four strains of mon-oclonal antibodies which could against H1N1,H3N2 virus and influenza B virus Yamagata lineage have been selected preliminarily by microneutralization exper-iment.These antibodies may have broadly neutralizing activity.Specifica lly the neutralizing activity of those monoclonal antibodies could be assayed With microneutralization.Finally the hemagglutination inhibition activity for th-ose antibodies could be detected through hemagglutination inhibition test. |
