Bangga herb, stemmed from Aconitum genus of Ranunculaceae family and with bitter in taste, cool in nature, and slight toxic, is one of important traditional Tibetan herb and frequently used for its various pharmacological actions in folk and clinical applications, such as anti-inflammatory effect, clearing away heat and toxic material, and tumor treatment etc. According to reported literature, Bangga herb, contained in many Tibetan compounds, mainly contain alkaloids which are its main active and toxic ingredients and reason for wide use. However, the quality standard of Bangga herb, extremely imperfect and lack of scientific and systematic, are greatly restricted the safe clinical medication.By conducted the extraction, separation, purification, and identification of alkaloids from Tibetan herb Bangga, three alkaloids named hordenine, heteratisine, and talatizamine were obtained, whose purity reach the requirements of the reference substance. By HPLC and TLC analysis, hordenine, with a common ingredient and significant pharmacological activities, was indicated as a evaluating index of Bangga herb.Consistent with literature reports, the qualitative analysis for toxic di-ester alkaloids by LC-MS has been found that Bangga herb dose not contains aconitine, mesaconitine, and hypaconitine at all. It is unnessary to establish a method of limited determination.In the study with hordenine as a detect index, both TLC method of qualitative analysis and HPLC method of quantitative analysis have been established to evaluate Bangga herb. According to determination of different batches, the content of hordenine was suggested not less than 0.009%. On the basis of the method of Chinese Pharmacopoeia (2015 edition), moisture, total ash, acid-insoluble ash, and extracts were studied in different batches and suggested not more than 9.5%,15.5%,2.5%, and not less than 23.4%, respectively.Established acute bronchitis model by fumigation, the pharmacological activity of alkaloids from Bangga herb was studied and found that these three alkaloids, hordeine, heteratisine, and talatizamine, have anti-inflammatory activity, and among them hordenine is strongest.By improved and established characteristi, identification, inspection and determination of test items, studied the pharmacological activity of alkaloids from Bangga herb, the quality standard of Bangga herb established could provide to support for rational utilization of resources, reseach for Bangga herb. |
