Objective: Proper exercise training can improve the function of the myocardial tissue, and long-term excessive training leads to myocardial tissue, or even a series of sudden cardiac death and a series of symptoms.Movement caused by myocardial hypertrophy, along with the changes of myocardial structure, whether can cause myocardial endoplasmic reticulum stress and apoptosis is less reported.Therefore this study through the establishment of different training intensity and training cycle motion model of rats, determination of left ventricular myocardium of rats endoplasmic reticulum stress related protein GRP78, CHOP, Caspase-12 protein expression, sports training and the effect and mechanism of myocardial endoplasmic reticulum stress occurred.Methods:Fifty-two adult male SD rats were divided into 5 and 10 weeks groups.(1) 5 weeks exercise group: control group(n=6), Time constant speed group(n=8), Speed increasing group(n=8),Time increasing group(n=8).control group only normal cage breeding.Treadmill exercise, training for 5 days a week.Exercise group rats velocity 10 m/min, constant speed constant speed group 1 week gradually increased to 24 m/min, for 60 min, last 4 weeks speed and time remain the same. Speed increasing set speed increasing every week about 3 m/min, 5th-week to 24 m/min, for 60 min. Time duration increasing group training week 1 speed gradually increased to 24 m/min, exercise duration per week increased 30 min, 5th week duration increased to 150 min.10 weeks exercise group model: control group(n=6), Time constant speed group(n=8), Speed increasing group(n=8).10 weeks model sports group of the before five weeks training plan the same with 5 weeks exercise group model, Time constant speed group of 24 m/min, for 60 min, until the end of the 10 weeks training,Speed increasing group tenth weeks to reach the speed of 36m/min, duration of 60 min.The rats were weighed weekly and recorded, each model in the hours of training time after the end of 24 rats were dissected, weighed heart weight, left ventricular weight; the left ventricular tissue, using Western blot to detect GRP78, CHOP, Caspase-12 protein expression, heart coefficient of rats finally calculated left ventricular coefficient.Result: five weeks exercise group model:(1) Speed increase group, Time increasing group rats compared with control group body weight, coefficient of heart, there are significant difference coefficient of the left ventricle, and constant speed relatively quiet group there was no significant difference;(2) Time increasing group rats myocardial GRP78 increased significantly, but the CHOP, Caspase-12 protein was no significant difference(3) Time constant speed group, speed increasing group rats myocardial GRP78 is quieter, CHOP, Caspase-12 protein expression group there was no significant difference.ten weeks exercise group model:(1) Increasing speed group compared with the control group of rats body weight, coefficient of heart, there are significant difference coefficient of the left ventricle, and constant speed group compared with the control group no significant change;(2) Time constant speed, speed increasing group rats myocardial GRP78, CHOP, Caspase-12 protein expression, there was no significant difference compared with quiet group.Conclusion:In the five weeks of training, To extend the duration of endurance exercise training exercise may induce endoplasmic reticulum stress, but no significant relationship between the movement speed increases despite increased GRP78 protein to extend the time of training, but CHOP, Caspase-12 protein does not appear significant changes,The datas suggest that the degree of ER stress-induced apoptosis strength may not reach. Ten weeks of training at constant speed or speed increasing endurance training no significant change in GRP78 tip has not yet cause myocardial endoplasmic reticulum stress. Extend the movement duration of endoplasmic reticulum stress related protein expression mechanism still needs to be further research.
