Aster is a kind of plants that belongs to Callistephus Cass genus of compositae,which is much accounted by horticulture field at home and abroad.It is widely distributed in Liaoning,Jilin,Shanxi,Hebei,Shandong,Yunnan,Sichuan and other places.Its flowers can be used as a medicine which has bitter taste and is mild-natured.What has been found now is that it contains eight kinds of polyphenol antioxidant,and they are apigenin,celery glucoside,kaempferol,hyperoside,naringenin,quercetin,luteolin and Kaempferol-7-O-β-D-glucopyranoside.Aster polyphenols is a kind of important biological active substances,which has strong antioxidant property,the ability to scaveng free radicals,and other functions,it may has prevention and treatment effect to liver damage and so on,but the corresponding data have not been reported.Therefore,the purpose of this study is to explore the function of China aster plant polyphenols as active ingredient for the prevention and treatment of liver fibrosis,and studying aster polyphenol’s possible mechanism to ease liver fibrosis effect.As a natural green and healthy food,aster has potential possibility of the prevention and treatment of liver damage diseases such as liver fibrosis and so on,which has great potential value for the development of aster’s own potential and the prevention and cure of liver fibrosis.The results we found were showed as below:Animal modeling and administration:(1)Treatment experiment: 6 weeks male KM mice were randomly divided into blank control group(n=12),Liver fibrosis model group(n=12),Low dose Asteroid polyphenol treatment group(n=12),High dose of Juju polyphenol treatment group(n =12),EGCG positive control group(n=12).Subcutaneous injection of CCL4 twice a week to replicate liver fibrosis models.At the same time,the EGCG treatment group was given 50mg/L,100mg/L Juju polyphenols and 100mg/L EGCG drinking water.The experiment lasted 6 weeks.(2)Prevention experiment: 6 weeks male KM mice were randomly divided into blank control group,liver fibrosis model group,low dose Asteroid polyphenols prevention group 2,high dose Asteroid polyphenols prevention group 2,EGCG positive control group.The same group of four weeks before the corresponding dose of distilled water,Juju polyphenols,EGCG aqueous solution.In the fifth week except for the blank control group,the other groups were subcutaneously injected with CCL4,twice a week to replicate the liver fibrosis model.At the same time,in the low dose of Juju Polyphenol 1 and high dose of Juju polyphenols 1 group to stop drinking Juju polyphenols,instead of drinking distilled water.The experiment lasted for eight weeks.Results:(1)Analysis on relieve effect of aster polyphenol in model mice with hepatic fibrosis:To analyse relieve effect of aster polyphenol in model mice with hepatic fibrosis,we mainly do experimental analysis on index number of viscera,hepatic fibrosis signal enzyme GOT in serum,pathological section,liver inflammation and pivotal proteins in TGF-β/Smad signaling pathway.After the delivery cycle over,execute the mice and have the liver and spleen to do the comparison of index number of viscera.For mice which drank aster polyphenol solution,their index numbers of viscera are notably expressed;the consequence of testing viscera and GOT,GPT in liver indicates that by drinking aster polyphenol solution experimental mice could reduce model mice’s index number of liver injury.Simultaneously,the consequences of experimental analysis on pathological section,inflammatory factor and TGF-β/Smad signaling pathway also reflect that the solution,to some extent,has relieve effect of Aster polyphenol in model mice with hepatic fibrosis.(2)analysis of mechanism of aster polyphenol easing fibrosis mice: TGF-β1 is the key factor to enhance HSC’s activation and promote liver tissue extracellular matrix(ECM).TGF-β1 can promote the synthesis of proteoglycan and collagen,elastin,adhesive protein such as extracellular matrix by HSC,thereby reducing the degradation of protease synthesis,preventing the new synthesis of ECM decomposition,breaking the balance between the ECM degradation and synthesis,and adding to the extracellular matrix deposition,thus accelerating the development of liver fibrosis.main function of TGF-β1 is delivering signals through the downstreaml Smads family to promote fibrosis,so TGF-β/Smad signal pathway plays an important role in the liver fibrosis mechanism.However the adjustion of plant polyphenols to TGF-β/Smad signal pathways mainly through inhibition of phosphorylation of receptor activation Smad and block of TGF-β/Smad signal pathways,achieving the liver protective effect.According to the results,what is found in the mice with liver fibrosis induced by CCl4 is that aster polyphenols can inhibit the expression of TGFβ1,Smad2,P-ERK1,P-JNK1,etc in TGF-β/Smad pathway.In the treatment group,effect of giving 50% of aster polyphenols is better than that of giving 100% of aster polyphenols.This research aims to rationally develop and make use of aster polyphenol as feasible hepatic fibrosis drug and explore effect of aster polyphenol acting as solution by drinking water to prevent and promote hepatic fibrosis;provide with statistical support on fully adjust effect of aster polyphenol on hepatic fibrosis bodies;do research on pivotal target-growth factor β1 and explore the mechanism of aster polyphenol preventing and promoting hepatic fibrosis to provide theoretical support for preventing treatment of hepatic fibrosis.The consequence of experiment shows that by given aster polyphenol,the symptom of hepatic fibrosis model mice induced by carbon tetrachloride can be relieved and this effect may be caused by stimulating related proteins in TGF-β/Smad signaling pathway in pancreatic and hepatic fibrosis. |
