| Objective:To investigate Allele frequency,genotype frequency,haplotype and such kind of relative population’s genetic polymorphisms of 12 X chromosome STR loci(DXS7132、DXS7423、DXS8378、DXS10074、DXS10079、DXS10101、DXS10103、DXS10134、DXS10135、DXS10146、DXS10148 and HPRTB)in Yanbian Korean population.What’s more,the objective of this paper is to confirm the application of forensic medicine.The study of parternity tests,personal identification and population genetics for Chinese Korean ethnic groups in China is based on the basic data.Evaluate the practical effectiveness of Investigator Argus X-12 kit of the research that X-STR of YanBian Korean.Methods:Sample collections:collect venous blood sample of 332 independent and healthy persons among whom the females are 176 and the males are 156 of Yanbian Korean population.Use Chelex-100 to extract DNA and then reaction apparatus to enlarge 12 X chromosome STR loci that Investigator Argus X-12 Kit handbook introduces.Apply 310 genetic analyzer to judge capillary electrophoresis and Alleles.Use Power Stats Software to analyze the data and acquire Allele frequency,heterozygosity,power of discrimination,polymorphism information content,and so on.The Genalex6.5 software is used to measure the frequency of the female population data,and was tested by the Hardy Weinberg balance(p-value).For the results of male,use the Arlequin software for haplotype diversity(HD),and use POPTREE2 software to analyze the group genetic data.Results:(1)Test out 6、6、7、19、9、18、25、4、20、9、5、23 Alleles in Yanbian Korean 12 X chromosome STR loci.For the 176 females,the result is that heterozygosity,H>0.5511,polymorphism information content,PIC>0.4306,power of discrimination,PD>0.6667 and all of the 9 Allele frequency accord with Hardy-Weinberg’ s law of equilibrium(P>0.05.For the males,test out 155 kinds of haplotype in the 156 independent persons.The most common haplotype is 16-10-15-39-17-32-21-14-27-19-14-20 and the polymorphism of haplotype is 0.9999.The attribution of polymorphism information content is 0.4306-0.9139.The attribution of heterozygosity is 0.5511-0.9318.Except DXS7423,the rest shows higher inherited characteristics.The results of Hardy-Weinberg(p-value)are over 0.05 except multiple alleles’ s DXS10134,DXS10135,DXS10148.Genetic data from the group shows that the genetic distance between the Yanbian Korean and the Koreans is the smallest,at 0.004,and the distance from the central Croatian is the longest,at 0.042.Conclusion:12 X-chromosome STR loci shows higher genetic statistics except DXS7423.These data are appropriate for forensic identification and the population genetics studies of Chinese Korean population.This research can provide fundamental material for the studies of Chinese Korean population’s paternity test,power of discrimination and population genetics.Investigator Argus X-12 kit is practical in the research of that X-STR of Yanbian Korean.The kit can be used in areas such as group genetics research and forensic application. |
