ObjectiveSpatial learning and memory impairment is the main clinical manifestation or chronic complication of many nervous system and non nervous system diseases.Scientific evaluation of spatial learning and memory behavior is a necessary and critical step in the development of basic research and development of new drugs.This study is based on the multi functional closed maze developed by the team of Liaoning Provincial Key Laboratory of behavioral and cognitive neuroscience,The aim of this paper is to establish the method of spatial learning and memory behavior evaluation,MethodEstablishment of spatial learning and memory behavior evaluation method based on multi functional closed maze.Through the preliminary experiment,we compared the different conditions of stimulation(sound stimulation,light stimulation,needle stimulation,acousto-optic stimulation),Daily training time(10 minutes,15 minutes,30 minutes),Training cycle(3 days,5 days,7 days),The average correct rate of the rats came,to determine the function of closed maze optimal stimulation conditions and optimal training and testing time.ResultEstablishment of spatial learning and memory behavior evaluation method based on multi functional closed maze.Through constant exploration,The optimum conditions of stimulation and stimulus intensity,i.e.the sound light water compound stimulus;Determine the best training test time,that is,10 min per day,training for 4 days,the test of fifth days,The trajectories of rats in the test area were recorded and analyzed.By optimizing the system,the combination of Noulds EthoVision XT11.5 and multifunctional closed maze,The automatic tracking and analysis platform for animal movement was established,and the regional/path behavior evaluation index was set up to reflect the spatial learning and memory ability of the experimental animals.The correct rate of latency,including water through the effective frequency,number of trial and error,the first time it target latency,sector retention time,movement distance,and the target sector and the average correct rate of the door.The application of spatial learning and memory behavior evaluation method based on multi functional closed maze.The accuracy and reliability of the proposed method are verified by the study of two animal models of cognitive impairment.Comparison with traditional Morris water maze evaluation methods,The results are consistent,and overcome the shortcomings,behavioral evaluation is more comprehensive.In D-galactose and alchlor compound induced animal model of chronic cognitive impairment,Through the evaluation of the new method,it was found for the first time that the model rats had depression tendency,The results were verified by forced swimming test.In animal model of acute cognitive impairment induced by scopolamine,The evaluation of the new method showed that the model rats had the tendency of delirium,which was consistent with that reported in the literature.The results of the present study support the changes of spatial learning and memory deficits in the two models at the cellular and molecular level.ConclusionIn this study,a new method of spatial learning and memory behavior evaluation was established based on the self-developed closed maze,The accuracy of the method was verified by two animal models of cognitive impairment.Compared with the traditional method,evaluation method of the evaluation index system has more advantages,rich and comprehensive,is conducive to improve the accuracy of evaluation and reproducibility of the results,is expected to provide a new platform for the research of neuroscience.
