| Background: Leprosy is a chronic infectious disease caused by Mycobacterium leprae.Leprosy affects the skin and peripheral nerves and can cause irreversible impairment on nerve function and chronic disability.Interleukin-27(IL-27)is a multifaceted cytokine with pronounced pro-and anti-inflammatory immunoregulatory functions.In 2015,Rosane and his colleagues found that IL-27 is upregulated in monocytes treated with IFN-β and Mycobacterium leprae,and the ability of IFN-β and M.leprae to induce IL-10 was diminished by koncking down of IL-27 in monocytes.These results showed a link between IL-27 and the IFN-β induced IL-10 pathway in leprosy,and suggested that IL-27 might contribute to the suppression of host antimicrobial responses in leprosy.Objective: To study the correlation between IL-27 and leprosy by measuring and comparing the expression of IL27 in leprosy patients and healthy controls.Methods: Measuring the expression level of IL27 in serum of leprosy patients and healthy controls by ELISA.Using quantitative PCR method to measure the expression level of IL27 in skin lesions of leprosy patients and that in the normal skin of healthy controls.Results: No significant difference in the expression of IL27 m RNA in peripheral blood was observed between leprosy patients and normal healthy controls.The concentration of serum IL27 in leprosy patients is obviously higher than that in healthy controls.The expressions of IL27 and genes in IL27 pathyway showed significant differences between skin lesions of leprosy patients and normal skin of healthy controls..Conclusion: IL27 and genes in related pathway showed certain correlation with the pathogenesis of leprosy. |
