| Background:The mortality of epithelial ovarian carcinoma is still highest among the gynecological cancer nowadays,although improved combination of surgery and chemotherapy is commonly applied..Paclitaxel plus platinum have become the standard first-line chemotherapy,which could make 70%patients achieve clinical complete remission.But most of patients would develop chemoresistance and eventually succumbing to their diseases.Acquired chemoresistant caused by multiple chemotherapy cycles has become a clinically formidable problem,which would cause treatment failure and death of ovarian carcinoma patients.Hence,it is important to explore chemoresi stant related factors for improving the prognosis of ovarian carcinoma.Objective:To detect the different expressions of CCNG1 and IL-8 in chemo-sensitivity or resistance tissues of epithelial ovarian carcinoma,and analyze the relationships between them and the influences on patients’ prognosis.Hope to provide a new target for reversal of drug resistance in ovarian carcinoma.Methods:196 cases of paraffin embedding tissues of ovarian carcinoma from January 2002 to May 2014 in Women’s hospital,School of Medicine,Zhejiang Univercity were retrospectively collected.The expression of CCNG1 and IL-8 were detected with the method of immunohistochemistry,and their relationships with the chemotherapy response status and the clinical pathological parameters of ovarian carcinoma were analyzed by chi-square test.The clinical pathological parameters include the pathological type(serous vs the serous carcinoma),pathologic stage(1,2vs3),FIGO stage(Ⅰ Ⅱ vsⅢ Ⅳ)and ovarian carcinoma "dualism" classification(type Ⅰ vs Ⅱ).The correlation of CCNG1 and IL-8 in ovarian carcinoma was analyzed by Gamma rank correlation.The significance of CCNG1 and IL-8 to the prognosis of ovarian carcinoma patients were analyzed with Kaplan-Meier,log-rank test.Furthermore,the fators associated with survival were analyzed using univariate and multivariate Cox Regression analysis.Results:1.The immunohistochemical results showed:the positive expression rate of CCNG1 in chemo-resistant ovarian carcinoma patients was 83.87%,which is significantly higher than chemo-sentitive group with 41.04%(P = 0.000);the positive expression rate of IL-8 in paclitaxel-resistant ovarian carcinoma patients was 90.32%,significantly higher than paclitaxel-sentitive group with 52.23%(P = 0.000).2.The relationship with clinical pathological parameters:Chi-square test showed the tendency that higher CCNG1 level is related with high grade and type II ovarian carcinoma,although the difference was not significant.And higher IL-8 level is related with late stage,serous carcinoma and type Ⅱ.3.Expression of CCNG1 and IL-8 in epithelial ovarian carcinoma were positively correlated(P = 0.000,rs = 0.312).4.Kaplan Meier and log-rank test showed that higher CCNG1 level indicated shorter overall survival and progress-free survival time of ovarian carcinoma,the difference was statistical significant.Similarly,higher IL-8 level indicated shorter overall survival and progress-free survival time of ovarian carcinoma,the difference was statistical significant.5.Univariate Cox regression analysis showed ovarian carcinoma patients’ overall survival and progression-free survival time were significantly associated with pathological types,histological grade,FIGO stage,ovarian carcinoma "dualism"classification(type Ⅰ-Ⅱ),the expression of CCNG1 and IL-8.The survival time of ovarian carcinoma patients became shorter when it was serous carcinoma,high grade,late stage,type Ⅱ ovarian carcinoma,high level of CCNG1 and IL-8.Multivariate Cox regression analysis showed that the expression of CCNG1 and FIGO stage were the independent factors affecting the prognosis of ovarian carcinoma patients.The expression of IL-8 is independent factor for progress-free survival time.Conclusions:The expressions of CCNG1 and IL-8 in chemoresistant ovarian carcinoma were significantly higher that of sentitive a tissue,which hints the higher expression of CCNG1 and IL-8 were probably related with chemoresistant ovarian carcinoma.Moreover,the expressions of CCNG1 and IL-8 were singnificantly associated with recurrence and death of ovarian carcinoma.So,CCNG1 and IL-8 may act as the markers which can predict the chemo-sensitivity and prognosis of ovarian carcinoma.In addition,the study discovered that the expression of CCNG1 was positively correlated with IL-8 in ovarian carcinoma,which suggested that CCNG1 might regulate the expression of IL-8.Further research is still needed to explore the intrinsic mechanism of chemo-sensitivity regulation in ovarian carcinoma.. |
