| Objective: To investigate the applicated value of dynamic contrast-enhanced M RI(DCE-MRI)in evaluating anti-angiogenic effect in human colon cancer transplanted subcutaneously in nude mice.Methods: 72 human colon cancer models of HT-29 which were made by nude mice were established,and divided into Avastin group 、KLT group and control group randomly,peritoneal cavities of the Avastin group were injected Avastin with dose 1.2mg/20 g one time,KLT group were injected KLT with dose 0.5mg/20 g per day and the control group were injected with the same volumes of saline in the same locations,DCE-MRI were performed before therapy and 2 days,7 days,14 days after treatment.8 mouses were sacrificed respectively after MRI scanning in three time points,and pathological immunohistochemical examination were done.The following parameters(Ktrans,Kep,Ve and Vp)which reflecting the tumor blood vessel functions were observed dynamically.The correlations between the changes of tumor tissue MVD 、α-SMA and parameter values at each time point were analyzed.Results: Ktrans and Kep of Avastin group and KLT group decline and the control group rise gradually with the extended of time,the statistical differences in the three groups were existed(P<0.05).There were no statistically difference between Ktrans and Kep of the three groups before treatments,however,Ktrans and Kep of the Avastin group and KLT group were obviously lower than the control group at each time point after treatments which were statistically significances(P<0.05).MVD counts of Avastin group were 9.8±0.6、7.8±0.5、5.6±0.7 respectively and α-SMA counts were 20.5±0.6、16.2±0.4、10.6±0.7,MV D counts of KLT group were10.2±0.4、8.3±0.6、6.0±0.4 respectively and α-SMA counts were 23.5±0.5、17.8±0.6、12.9±0.5 respectively,obviously lower than the control group MVD 13.6±0.4、16.4±0.7、20.4±0.9,α-SMA 29.5±0.7、35.8±0.9、46.9±1.0.Ktrans and Kep of the three groups after treatments were positively correlated with corresponded MVD and α-SMA at each time point(P<0.05).Conclusion: Avastin and KLT could inhibit the development of human colon cancer xenograft significantly,the dynamic changes of Ktrans and Kep values of DCE-MRI technology can be considered as a quantitative indexes for evaluating early colon cancer chemotherapy. |
