[Objective]To observe the probability of chlamydia protein Pgp3 inhibit the development of imiquimod(IMQ)-induced psoriasis-like inflammationon mice by combining with endogenous antimicrobial peptide LL-37. [Methods]The GST-Pgp3 protein was prepared by E.coli expression system and purified by glutathione S-transferase(GST) MagBeads. Pgp3 was cleavedfrom GST-Pgp3 by PreScission Protease. BALB/c female mice(n=32) were randomly divided into 8 groups: Control, Model, Ext-H, Ext-L, Ext-C, Inj-H, Inj-L, Inj-C. Thelesions on mice were evaluated according to the psoriasis area and severity index(PASI). The morphological changes of lesions were observed under light microscope and the epidermal thicknesses were measured by micrometer. [Results]Compared with model group, the erythema and scale in Pgp3 treated groups were alleviated, with lower PASI scores. Epidermal over-proliferation and epidermal parakeratosis were relived. Acanthosis and apodeme extension were descended. The changes of epidermal thicknesses were significant different in Ext-H group and Inj-H group comparing with model group and PBS treated groups(P﹤0.05). However there were no statistical significance in Ext-L group and Inj-L group comparing with model group and PBS treated groups(P﹥0.05).The lesions of PBS treated groups were similar to model group, and there were no significant changes in either morphological or epidermal thicknesses under light microscope(P﹥0.05). [Conclusion]Pgp3 could inhibit the development of IMQ-induced psoriasis-like skin inflammation, we conjecture that Pgp3 could combined with LL-37 to inhibit the psoriasis-like lesions on mice.
