Objective: There are many allergen detection method,The main method of detect the serum allergen special immunoglobulin E(lg E) in vitro was Immuno Cap,which was as the “gold standard”. Compared with the traditional solid support, the cellulose derivatives with more protein adsorption and faster reaction speed advantaged as faster reaction speed, higher sensititivity and specificity; however the price is relatively higher. The solid support of Western Blotting is nitrocellulose membrane,which was poor sensitivity and sepecificity. In recent years, Liquichip technology was widely recognized and performed in the field of medicalscience. We developed the Liquihip method to detect the serum allergen specific Ig E levels, comparing with the widely used Western Blotting in clincal, to evaluate the Liquichip method in the linear, sensitivity, specificity, and other indicator. We study a new strategy for the clinical diagnosis of allergies.Methods: 120 cases of serum spicemens of allergic patients was collected in the department of dermato Logy, Tianjin Academy of Traditional Chinese Medicine Affiliated Hospitalin from Oct.1 to Dec.1.2015. Western blotting was prefromed to detect serum allergen specific Ig E levels. The positive rate of serum allergen specific Ig E was analysed and the research objects was selected as the control group.Liquichip method was used to detect the serum level of dust mite allergen specific Ig E levels and the relationship of the two methods was analyzed via linear correlation.Liquichip method was used to detect the milk serum allergen specific Ig E and mugwort serum allergen specific Ig E levels, and to evaluate its clinical compliance.We use SPSS18.0 to analysis the variables. To compare s Ig E levels among two groups with non parametric test. Using Spearman rank correlation coefficient to evaluate the level of correlation. To select Wilcoxon rank sum test to compare paired data, P < 0.05 was considered as statistically significant.Results:Western blotting was used to detect serum allergen specific Ig E levels in 120 cases of allergic patients. Using Liquichip technology, the positive rate of mite allergen specific Ig E is up to 61.67%; the linear range of the standard curve of tota L Ig E is 0.41ng/m L~100ng/m L. In order to test repeatablity. using Liquichip method to detect the serum tota L Ig E levels in 10 allergic patients,the intra- and inter- assay coefficient of variation(CV) are 1.20%~2.86% and 1.84%~4.91% respectively. As Western blotting, we use Liquichip method to detect serum mite allergen specific Ig E levels in 50 patients who are randomly selected from 120 patients in accordance with grade. r=0.55(P < 0.001, n=50), showed that the two methods of detection of serum allergen specific Ig E levels are significant Ly correlated. Using Liquichip method respectively to detect the allergen specific Ig E levels in 10 patients who have been diagnosed as milk allergy and in 10 patients who have been diagnosed as mugwort allergy. the results showed that the positive rate is 100%, with good clinical compliance.Conclusion:1. The highest positiverate of dust mite was detected with traditional Western Blotting, followed by cockroaches, soybeans, milk and mugwort2. We developed Liquichip method with Biominda patent to detect total Ig E and specific Ig E, which has wide linear range, good repeatability, and stable results.3. The correlation between Liquichip method and Western Blotting was good; the sensitivity of the former was better.4. We developed Liquichip method to detect serum milk allergen specific Ig E and mugwort allergen specific Ig E, and clinical compliance was 100%, Sensitivity was higher.
