| Objective:Screening and verification lncRNA expression profiles associated with colorectal cancer and making preliminary bioinformatic analys is of differentially expressed mRNAs.Methods : Selected four cases of colorectal cancer tissues, extracting RNA,conducting microarray hybridization. GO analysis and KEGG analysisof differentially expressed mRNA.GO classify into biological processes 、 cellular components and molecular functions based on Gene Ontology.A significant degree of GO functional pathways by P value indicates that the more significant functional ways with the smaller P value; A significant degree of KEGG biological pathways by P value indicates that the more significant biological path ways with the smaller P value;Co-expression network:Gene co-expression networks were built according to the normalized signal intensity of specifically expressed genes.Result:The differential expression data of lncRNAs was 3523(1989 lncRNAs were up, 1534 lncRNAs were down),the differential expression data of mRNA swas2515(1471 mRNAs were up, 1044 mRNAs were down);GO analysis showed that mRNAs differentially expression mainly enriched in Biologys 、 components and molecular functions, the highest enrichment degree of biological process was the mitotic cell cycle in upregulated mRNAs; the highest enrichment degree of cell component was theon-membrane-bound organelles in upregulated mRNAs; the highest enrichment degree of molecular functiont was the protein binding in upregulated mRNAs;the highest enrichment degree of biological process was the organic hydroxyl compound metabolic processes in downregulated mRNAs; the highest enrichment degree of cell component was the extracellular region in downregulated mRNAs;the highest enrichment degree of molecular functiont was the steroid binding in downregulated mRNAs;KEGG analysis showed that mRNAs differentially expressed mainly concentrated in the twenty-two signaling pathways, these mRNAs may play a important role in biological pathway. The highest enrichment degree of biological pathway was the hsa05322(Systemic lupus erythematosus-Homo sapiens) in upregulated mRNAs.The highest enrichment degree of biological pathway was the Systemic lupus erythematosus- Homo sapiens(human) in downregulated mRNAs.moreover the two biological pathway:Wnt signaling pathway’was previously identified as a crucial initiation step of colorectal Cancer and Retinol metabolism has been reported to be involved in colorectal cancer metastatic multiplicity.As mentioned above, the ‘Wnt signaling pathway’ was upregulated in CRC tissues compared to non-cancerous tissues.coding-non-coding gene co-expression network(CNC network) was constructed between the top 10 differentially expressed ‘Wnt signaling pathway’ genes and all differentially expressed lncRNAs.Conclusion : Microarray datashowed that a total of 3,523 lncRNAs and 2,515 mRNAs ere consistently differentially expressed in the CRC tissues compared to adjacent normal tissues; Coding-non-coding gene co-expression nalysis showed that these differentially expressed lncRNAs ere closely correlated with ‘Wnt signaling pathway’ compo-nents, whose aberrant activation plays a central role in CRC. |
