s.mutans is the main pathogenic bacteria of dental caries, which is the most common infectious oral disease. Therefore, inhibiting the proliferation of s. mutans has great significance of controlling the development of caries disease. STAMPs are composed of the killing regions, the targeting regions and/or linker regions. It can enhance the antimicrobial activity by increasing the targeting regions binding to the surface of the targeted pathogen. LL-37 is the only α-helical structure Cathelicidins peptides that has been founded so far. LL-37 has strong antibacterial activity, high stability, and it leads to pathogen mutated rarely. So LL-37 is the ideal template synthesis and molecular skeleton for the designing and synthesising of new antimicrobial peptides. In addition, many scholars have synthesised the STAMPs successfully by utilizing CSPC16(S. mutans competence stimulating peptide) as the killing regions. So that, the objective of this artical is to synthesis an specifically targeted antimicrobial peptides C16LL-37,and the killing regions is CSPC16, the targeting regions is LL-37, the linker regions-GGG-. At the same time, to evaluate the biological characteristics of C16LL-37.Methods: In this study, an antimicrobial peptide LL-37, a peptide derived from CSPC16(S.mutans competence stimulating peptide) and recombined peptide C16LL-37 were synthesized by Fmoc-chemistry-based strategy. The selectivity and antibacterial activity of C16LL-37 were identified by colony counting method on microbiobial culture plate. After treatment of C16LL-37 at 32μM, the morphological changes of S. mutans were observed by using SEM.Results: 1. The minimum inhibitory concentration of C16LL-37 was 16μM, and the minimum bactericidal concentration was 64μM; 2.The survival rate of S. mutans was 3.46% after C16LL-37 treated at 64μM for 30 mins, while it was 0% at 64μM for 60 mins. The survival rate of other four kinds of bacteria, however, were more than 60% at any time(P<0.05); 3. The morphological change of S. mutans was observed after C16LL-37 treatment at 64μM by using SEM, which presented irregular in shape, and rough surface. The S. mutans furthermore became splitting.Conclusion: C16LL-37 has obvious specificity for S. mutans, strong antibacterial activity.These characteristics of C16LL-37 implies good perspective in caries research and clinical application. |
