Aim We used DAPT intervention in human gastric cancer cell lines BGC-823.we explored the relationship between the expression of Notch1 and MMP-9,as well as the mechanism of Notch1 may involvement in the invasion and migration of gastric cancer.Method After the treatment of DAPT in different concentration at different time points on cultured human gastric cancer cell lines BGC-823,the MTT method was employed to detect survival rate of the cells.The real-time quantitative PCR was used to detect the level of MMP-9mRNA respectively.Western blot was used to detect the expression of Notch 1 intracellular domain NICD and MMP-9 protein.Result①DAPT inhibited the growth of human gastric cancer cell lines BGC-823 in vitro.②The expression of MMP-9mRNA declined and performed a dose-time dependent manner after intervention of DAPT;③DAPT inhibited the expression of Notchl intracellular domain NICD and MMP-9 protein Notchl protein and performed a dose-time dependent manner.④The expression of Notch1 and MMP-9 showed a positive correlation(r =0.575,P<0.05).Conclusion DAPT may inhibit the expression of Notchl,down-regulate the expression of MMP-9.Our results confirm that Notchl involved in tumor invasion and migration process by regulating the expression of MMP-9.
