Objective: To Explore the impact of COPD rats skeletal muscle function and its’ expression of FOXO1 in different doses of salidroside. To provide an experimental evidence of salidroside used in the treatment of the improvement of skeletal muscle dysfunction patients with COPD.Method: 40 adult SD rats were divided into 5 groups(each group of 8),respectively is: the blank group, COPD model+ placebo(CS), COPD model + low-dose salidroside group(CS + L), COPD model + mid-dose group(CS + M),and COPD model+high-dose salidroside group(CS + H). Firstly, COPD model+ placebo(CS), COPD model + low-dose salidroside group(CS + L), COPD model + mid-dose group(CS + M),and COPD model+high-dose salidroside group(CS + H) all adopts pure passive smoking smoking rat model was established, blank group take false exposure method to establish the model of the blank group. Blank group and(CS) given a placebo for 1 month.COPD model+ placebo(CS), COPD model + low-dose salidroside group(CS + L),COPD model + mid-dose group(CS + M),and COPD model+high-dose salidroside group(CS + H) respectively, to low dose(5 mg / 100 g), middle dose(25 mg / 100 g), high dose(125 mg / 100 g) salidroside lavage treatment 1 month. Record its weight changes, the use of trying to swim experiment assessment exercise capacity, serum, lung tissue and quadriceps, determination of skeletal muscle samples determination of type I fiber proportion, FOXO1, P-, Atrogin-1 FOXO-1, Mu RF-1 level.Results:(1) Wight cmpasion: CS group(404.6 + 31.7 g) compared with group C(475.6± 20.4 g), the difference was statistically significant(P < 0.05); CS + L group(419.3±20.9 g), CS + M group(425.6± 32.6 g), CS + H group(432.7±27.2 g) compared with CS group, weight increased, and the value between the difference had statistical significance(P < 0.05).(2)Forced swimming test(FST) comparison: CS group(52.4±4.5 min) compared with group C(14.9±4.1 min), the difference was statistically significant(P < 0.05); CS + L group(22.5±2.9 min), CS + M group(23.1±2.9 min),and CS + H group(31.1±4.7 min) compared with CS group, have extended swimming time, and each value between the difference had statistical significance(P < 0.05).(3)The quadriceps weight comparison: CS group(2.82± 0.22 g) compared with group C(2.82±0.22 g), the difference was statistically significant(P < 0.05); CS + L group(2.99±0.20 g), CS + M group(3.13± 0.23 g),and CS + H group(3.44± 0.23 g) compared with CS group,the weight of quadriceps were increased, and each value between the difference had statistical significance(P < 0.05).(4)The proportion of type I muscle fibers camparison: CS group(16.3±4.9%) than group C(32.8±2.8%), the difference was statistically significant(P < 0.05); CS + L group(22.2±3.8%), the CS + M group(25.7±3.9%), CS + H group(28.2±2.9%) compared with CS group, rising rates of type I muscle fibers, and each value between the difference had statistical significance(P < 0.05).(5) Immunohistochemistry result:(1)FOXO1: group C(0.260±0.10), the CS group(0.646 ± 0.11), the CS + L group(0.440 ± 0.13), the CS ±M group(0.380± 0.12), CS + H group(0.330± 0.01);(2)P- FOXO1: group C(0.230±0.06), the CS group(0.343 ± 0.08), the CS + L group(0.401± 0.06), the CS + M group(0.314 ± 0.07), CS + H group(0.289 ± 0.06);(3)Mu RF- 1: group C(0.338±0.077), the CS group(0.634± 0.101), the CS + L group(0.428 ± 0.067), the CS + M group(0.530 ± 0.065), CS + H group(0.353 ± 0.105);(4): Atrogin- 1 C(0.393±0.013), CS group(0.370±0.071), CS + L group(0.306 + 0.103), the CS + M group(0.286 + 0.005), CS + H group(0.157±0.061). Compared with CS group, increased FOXO1 expression, and the value between the difference had statistical significance(P < 0.05). CS group compared with group C FOXO1, FOXO1, P- FOXO, Mu RF- 1, expression levels, differences were statistically significant(P < 0.05); CS + L group, the CS + M group, the CS + H group compared with CS group, FOXO1, FOXO1, P-, Atrogin FOXO, Mu RF- 1-1 of expression level decreased, and the influence and intervention levels were positively correlated, the difference had statistical significance(P < 0.05).Conclusion:. Salidrosid may be through cut FOXO-1 expression to ease COPD model rat skeletal muscle dysfunction,and the protection were dose correlated.
