| Objective:To investigate the effect of insulin and glucose on the expression of s ES,MMP-9 and TIMP-1.Methods:Primary cultured smooth muscle cells from the internal carotid artery and endothelial cells from the aorta of SD rats treated with different doses of insulin and glucose, s ES,MMP-9 and TIMP-1 released by cells into medium were quantified by ELISA.Results:s ES and MMP-9 levels were higher in insulin group compared with control(no insulin) group(P<0.05 and P<0.05),while TIMP-1 was less than control group(P<0.05). Significant positive correlation were observed between insulin and s ES(r=0.841,P<0.001)as well as MMP-9(r=0.815,P<0.001),negative correlation were observed between insulinand TIMP-1(r=-0.740,P<0.001).s ES and MMP-9 levels were higher in glucose group compared with control(no glucose) group(P<0.005 and P<0.001),while TIMP-1 was less than control group(P<0.001). Significant positive correlation were observed between glucose and s ES(r=0.972,P <0.001)as well as MMP-9(r=0.973,P<0.001), negative correlation were observed between insulin and TIMP-1(r=-0.963,P<0.001).Conclusion:Insulin and glucose in a range of dose augmented the secretion of s ES and MMP-9, attenuated the secretion of TIMP-1. |
