Objective: This study was to explore the plasma VEGFA level change and VEGFA SNPs(Single Nucleotide Polymorph Sims) allele distributions and differences between hainan li and han normal people and COPD patients; Reveal whether SNP in VEGFA gene were susceptibility gene in hainan li and han nationality patients with COPD; Clarify the correlationship between VEGFA gene polymorphism, The level of plasma VEGFA and COPD Risk.Methods: I In this case-control study, we recruited 474 subjects of li ethnic(COPD 234, Normal control 240), han ethnic included 596 subjects(COPD 246, Normal control 350). Collected Peripheral blood and extraction of genomic DNA, we genotyped 9 SNPS(VEGFA gene) by the Mass Array and using the x2 test and genetic model analysis evaluated their association with COPD. SNP genotyping was done in 495 patients(249 COPD patients and 246 control group), which were randomly selected plasma specimen from the VEGFA genotyping of SNP has been done. ELISA method was used to detect the level of VEGFA in plasma. According to the layers of nation, gender and smoking status, the expression difference was analyzed between case and control, and further analysis the correlations between VEGFA gene polymorphism and plasma VEGFA levels in the hainan li and the han population. Further analysis the correlation between VEGFA SNPs and its protein and COPD risk factors such as smoking etc.Results:The case control study found that:(1)rs833068,rs3024994,rs3024997 and rs3025000 loci allele frequencies of VEGFA were significantly different among Li and Han population(P<0.05).(2)VEGFA gene rs3025030(OR=1.40, 95%CI=1.05-1.86, p=0.027) and rs3025033(OR=1.37, 95%CI=1.03-1.82, p=0.036) Correlation with COPD susceptibility in Hainan Han population, While may not be related to Li population.(3) The plasma VEGFA level of Li population both in COPD and control were lower than that in Han population Corresponding grouping(P<0.05).(4)The factors Increased expression of plasma VEGFA in Li population: high BMI, advanced age, duration of smoking, Chronic obstructive pulmonary disease(COPD), recurrent respiratory tract infection, Biofuels, Frequently induced cough, Family history of respiratory diseases, recent respiratory tract infection, passive smoking and COPD exacerbation.(5)The factors Increased expression of plasma VEGFA in Han Population: duration of smoking, COPD, low educational level, recurrent respiratory tract infection, Frequently induced cough, Family history of respiratory diseases, recent respiratory tract infection, COPD exacerbation.(6)The plasma VEGFA levels of COPD in acute exacerbation phase were higher than that in remission stage and control group, the plasma level of VEGFA was lower in COPD than in control group.(7)We found s3025030 and rs3025033 in VEGFA gene which increase the risk of COPD also associated with plasma VEGFA levels. Both Rs3025030 and rs3025033 minimum allele can increase the plasma VEGFA level. We also get a similar conclusion according to stratified analysis by smoking status, COPD incidence and ethnic.(8) Multiple linear regression analysis showed that the Li population plasma VEGFA levels related to BMI, grouping, recent respiratory tract infection,biofuels and family history of respiratory disease. In the Han population, plasma VEGFA was associated with duration of smoking, recent respiratory tract infection and Rs3025030.Coclusions:(1)rs833068, rs3024994, rs3024997 and rs3025000 loci allele frequencies of VEGFA have genetic heterogeneity among Li and Han population(P<0.05).(2)rs3025030 and rs3025033 were susceptibility loci of COPD in Hainan Han population.(3)The expression level of VEGFA in plasma was different between Li and Han population.(4) The expression level of VEGFA in plasma was different in different periods of COPD.(5) The expression level of plasma VEGFA both in Li and Han population were positively correlated with duration of smoking.(6) VEGFA gene Rs3025030 and rs3025033 loci minimum allele which increase the risk of COPD also associated with High expression of plasma VEGFA levels.(7) The influence factors of plasma VEGFA level in Li and Han population were different.
