| One type of ulcerative colitis(UC) belonging to inflammatory bowel disease(IBD), and lesions in the mucosa and submucosa, mainly involving Ulcerative colitis(UC) is one type of inflammatory bowel disease(IBD),lesions were located at the mucosa and submucosa, mainly involving the colon and rectum. The pathogenesis of UC is not yet entirely clear. The reason of UC was main due to the environmental factors, genetic immunization, and pathogenic microorganisms, respectively. The disease can harm gastrointestinal track as well as other organs. Turner-Warwick M et al. firstly reported that UC can lead to respiratory diseases. And Kraft SC found that pulmonary lesions was parenteral performance. In the study, the acute phase UC animal model induced by 4% dextran sulfate sodium was conducted by male BALB/c mice which were free to drink 4% dextran sulfate sodium(DSS). After intervention of compound dracorhodin enema,immunohistochemical,the expression quantity of interleukin-6(IL-6) in colon and lung were measured by histological scores, ELISA, RT-QPCR assay. This was as the index of judging prognosis and curative effect, which provide experimental basis for compound dracorhodin enema alleviating UC and UC lung injury.To explore the protective effect of compound dracorhodin enema to colon and lung tissue of mice UC model, 60 male BALB / c mice were randomly divided into four groups viz. normal group, DSS group,Dracorhodin group, DSS + Dracorhodin group, each group has 15 mice. All the 60 mice were feed 7 days to adapt to the environment. The eighth day,the normal group were gavaged with 1ml normal saline; DSS group were given 4% DSS as well as 1ml normal saline; 1ml Compound DracorhodinResin(0.075 g / kg / d) was given to the dracorhodin group. DSS +Dracorhodin group was 4%DSS as well as 1ml Compound Dracorhodin Resin(0.075 g / kg / d). 10 days later, the general condition, disease activity index(DAI) and histological evaluation of tissue severity score of inflammation was observed. The number of positive cells were observed by immunohistochemistry, the IL-6 expression in colon and lung tissue was measured using ELISA and RT-QPCR assay.Results showed that there is no obvious inflammatory performance in the normal group and dracorhodin group; DSS group and DSS +Dracorhodin group have severe and mild inflammatory performance. The DAI scores were significantly increased in both DSS group and DSS +Dracorhodin group, but the DAI score task back in DSS + Dracorhodin group and there are significantly difference between the two groups(P<0.05); the colon length were( 5.220± 0.405) cm,(6.380 ± 0.434) cm in DSS group, DSS + Dracorhodin group, respectively. Colon histological(HI)scores were(6.150 ± 0.145),(2.640 ± 0.056), group lung histology scores were(7.320 ± 0.079),(3.360 ± 0.157) in the two groups. Result showed there are significantly difference between the two groups(P<0.05);IL-6expression by immune histochemical method was used to detect colon and lung tissue under the microscope observation, the number of positive cells in DSS + Dracorhodin group was significantly less than in DSS group(P<0.05); ELISA was used to detect IL-6 expression in colon and lung tissue,The expression quantity was(66.511 ± 1.171,76.068 ± 0.537) pg/mg, and(19.121 ± 3.141,24.791 ± 2.332) pg/mg in DSS group and DSS + IL-6group, respectively, which also showed significant difference(P <0.05)between two groups. The expression quantity of messenger ribonucleic acid(m RNA)in colon and lung tissue was measured using real-time quantitative polymerase chain reaction(RT-QPCR), the expression quantity is14.087±2.143, and 6.536±2.432 in colon and 15.971±0.170 and8.042±0.671 in lung tissue in DSS group, DSS + Dracorhodin group, whichshowed significant difference in the two groups(P<0.05).In present study, mice UC model was successfully conducted by gavaging with DSS, which was similar with human UC symptoms and clinical manifestations. By clinical symptoms in each group of mice, DSS +Dracorhodin group was more severe than normal mice, and milder than model group according to the general situation, DAI scores, histological scores. The results of IL-6 expression by ELISA, immunohistochemistry,RT-QPCR assay in colon and lung tissue showed that compound dracorhodin enema have good effect on UC in the mice’s lung and colon,which provide strong experimental theory basis for the viewpoint“compound dracorhodin enema can reduce the expression of IL-6, ease UC and UC lung injury in mice”. The results brings new hope for the treatment of UC and UC lung injury. |
