| Objective: To observation of of Phlegm Kiyokata blood lipid induced in TC,TG,LDL-C,HDL-C levels and the t-PA,PAI-1 effect of interventions to further explore living phlegm stasis phlegm and clear party the mechanism for the clinical treatment of hyperlipidemia and improve fibrinolytic activity,protect endothelial function provides a scientific basis.Methods: 60 SD rats were randomly divided into control group,model group,high dose group,middle dose group of traditional Chinese medicine,Chinese medicine low dose group and western medicine group,a total of six groups,each group 10,control group given normal diet,the rest of the group was given high-fat diet.5 weeks,the control group and model group were given normal saline(2ml/100g?d)orally,according to traditional Chinese medicine high dose group(9g / 100g?d),middle dose group(1.8g/100g),low dose group(0.9g/100g),respectively,given the high,medium and low concentrations of phlegm and clear granules(2ml/100g)orally,Western medicine group were given atorvastatin calcium(1mg/100g?d)gavage.The first end of 8 weeks,all rats were sacrificed,the abdominal aortic blood,serum markers measured in all the groups TC,TG,LDL-C,HDL-C and plasma indicators t-PA,PAI-1 and liver and kidney function(ALT,AST,BUN,Cr);the general case,the recording was observed throughout the experiment all rats mental state,food,urine,hair and body weight changes.All data were statistically analyzed using SPSS19.0 software,the difference expressed as mean ± standard((?)±s),in order to test the level of α=0.05Results: Before 1 gavage,the growth trend weight control group of rats was significantly lower than the other groups;gavage process,phlegm lofty,medium and low dose group and western medicine group body weight growth trends are slowing down,still the model group increased weight relatively fastest,weight control group had steady above results suggest that: compared with the normal diet,high fat diet on body weight change is more obvious level.2 serum lipid model group TC,LDL-C,TG levels were significantly higher than the control group,statistical comparisons between the two groups was significant difference(P<0.01),suggesting rats hyperlipidemia.3 Western medicine group compared with the model group,TC,LDL-C,TG levels decreased significantly(P<0.01);Phlegm medicine Qing group compared with the model group,high dose group and middle dose group TC,TG,LDL-C level and low-dose group TG levels were significantly lower(P<0.05),while the low dose group TC,there was no significant difference(P>0.05);each group HDL-C was no significant difference(P> 0.05).These results suggest that: atorvastatin calcium intervention on plasma lipid levels in the most obvious,and traditional Chinese medicine phlegm and aloof,middle dose group on blood lipid levels can also play a role in the intervention,but the effect is not atorvastatin calcium effect is obvious.4 Comparison with the control group,serum PAI-1 was significantly increased in the model group(P<0.01)and t-PA is decreased significantly(P<0.01),model group reduces prompt fibrinolytic activity levels in rats,hypercoagulable tendencies;compared with the model group was significantly increased(P<0.01)dose serum t-PA,PAI-1 decreased significantly(P<0.01),low-dose group,t-PA was significantly higher(P<0.05),PAI-1 decreased significantly(P<0.05),while the western group,high dose group compared with the model group was not statistically significant(P>0.05),phlegm and clear description of the low-dose group could improve rats with fibrinolytic activity,and the western group and Phlegm lofty dose group and the model group had no improvement.5 Safety indicators comparison: During the experiment,the high-dose group and each group of death a rat,anatomy,analysis of the reasons may be improper operation gavage needle strayed into the trachea,filling juice into the lungs and death.Chinese medicine and Western medicine each dose group,no end of the experiment the model group,serum markers of ALT normal group,AST,BUN,Cr levels with the control group statistically significant difference(P>0.05),showed no abnormal liver and kidney function,clinical drug safety better;more than the growth of the state,diet,water intake were abnormal situation.Conclusion: 1 uses high-fat diet feeding SD rats,hyperlipidemia model rats replication is feasible;2 Chinese medicine phlegm and clear the same party can level of blood lipid in rats with TC,LDL-C,TG play a role in the intervention,but the effect is not as good as atorvastatin calcium effect is obvious;3 phlegm and clear,the low dose of t-PA can increase the activity of PAI-1 inhibited improved fibrinolytic activity,possibly by improving blood rheology,reduce cholesterol and other multi-target protection of endothelial function. |
