| Objective: This experiment by using Langendorff perfusion apparatus of rat isolated heart perfused with verapamil hydrochloride induced slow arrhythmia model method,with rhythm Corning extract intervention slow type arrhythmia isolated heart irrigation flow model in rats and observe the rhythm Xinlv Kang Ning on isolated heart slow slow type arrhythmia model in rats with heart rate(HR)and left ventricular pressure(LVSP)peak and explore rhythm Xinlv Kang Ning treatment of slow type arrhythmia(heart and kidney yang,Xinmaitong Bizu)at the same time,improve the mechanism of cardiac function,for traditional Chinese medicine in the treatment of slow arrhythmia provide experimental basis,research ideas and methodsMethods: A total of 30 SPF male healthy SD rats,weight 250-280 g,provided by the experimental animal center of Henan province.These rats were randomly divided into cardiac Xinlv Kang Ning extract low middle high dose group,model control group and salbutamol sulfate group,8 rats in each group.Using a Langendorff perfusion apparatus of isolated rat heart perfused with verapamil hydrochloride and induce slow arrhythmia model method,comparative study of rhythm Xinlv Kang Ning extract and salbutamol sulfate on isolated heart slow type arrhythmia model rat heart rate and left ventricular pressure peak level of improvement,and statistical analysis.Results: Xinlv Kang Ning(Low,medium and high dose group),salbutamol sulfate group and model group had significant difference compared with control(P<0.05).Xinlv Kang Ning high dose group and salbutamol sulfate group had significant difference(P<0.05).Xinlv Kang Ning medium dose group and salbutamol sulfate groups were not statistically significant(P>0.05).Xinlv Kang Ning low and middle dose group had significant difference(P<0.05Conclusion:(1)The results suggested that the method of using Langendorff perfusion device to induce the model of slow arrhythmia induced by Vera Pammy in isolated perfused rat hearts was feasible.(2)Xinlv Kang Ning can obviously improve isolated heart arrhythmia in rat model of heart rate,and the high dose of the best effective dose.(3)Xinlv Kang Ning can improve isolated heart slow left ventricular arrhythmia in rat model of internal pressure peak,high dose of the best effective dose(4)Xinlv Kang Ning can improve isolated heart slow left ventricular arrhythmia model in rats can increase the peak pressure level,myocardial contractility,and improve cardiac function of systolic function by regulating calcium ion channels or interacting with other ion channel target and improve the electrical activity of the heart in vitro and to provide experimental basis for objective. |
