| Objective: From the perspective of metabolic pathways of astrocyte glutamate, th e author preliminary studied the regulatory role of acupuncture on MCAO rats and on glutamate transporter1(GLT1) and glutamine synthetase(GS) which are the cyclekey links of glutamate- glutamine cycle,which provide new ideas and experiment al basis for the study of the mechanism of acupuncture treatment of ischemic strok e.Methods: we produced the middle cerebral artery occlusion(MCAO) using suture method, then we needle MCAO Rats Neiguan for 7 days, and let Ground hole machine as a relatively irrelevant points for comparison, use methods such as neural behavioral score,ischemic brain tissue morphology, biochemical detection and immunohistochemical detection, the we observed the recovery of MCAO rats neurological function, the content of Brain glutamate and glutamine synthetas, and the protein expression of Astrocytes GLT1 and GS.Preliminary discussion acupuncture on MCAO rats glutamate- glutamine cycle key glutamate transporter 1(GLT1) and glutamine synthetase(GS) regulation function.Results:1. Continuous acupuncture Neiguan for 7 days can significantly improve the damage of MCAO rat brain tissue, and also promote the recovery of neurological function.2. Acupuncture Neiguan can reduce the excessive glutamate on MCAO rat brain tissue;and increase the content of the brain tissue of glutamine synthetase, and improve the content of glutamine synthetasethe on brain tissue.3. Acupuncture Neiguan can improve the protein expression of GLT1 and GS in astrocytes.Conclusion: By increasing the expression of GS and GLT1 in astrocytes, acupuncture Neiguan can regulate the glutamate metabolic pathways of astrocytes, enhanced the ability to remove excessive glutamate, relieve neural excitotoxicity, and exert the protection and treatment for cerebral ischemia. |
