Objective:Researching the correlations between the Leptin activated PI3K/AKT signaling pathway and VEGF the expression levels of Hela Cells, and analysis of the molecular mechanisms.Methods: 1.The experiment includes four groups: Control group, Leptin Ⅰ group, Leptin Ⅱ group, Leptin Ⅲ group, Hela Cell Lines were incubated with 0 ng/ml,50ng/ml,100ng/ml and 250ng/ml Human recombinant Leptin for 0 h,12 h, 24 h, 48 h,72h and then measurement of OD value changes in 450 nm, analysis Hela Cells proliferation in different concentrations, Selected proliferation effect the best time point.2.At the best time point, Hela Cell Lines was treated by Human recombinant Leptin in different concentrations, observed their morphological of Hela Cells by microscope.3. At the best time point, Hela Cell Lines was treated by Human recombinant Leptin in different concentrations, the expression levels of p-AKT,HIF-1α and VEGF by Western Blot, and analysis differences.Results:1.CCK8 results show that Leptin in different concentrations can induced Hela Cells proliferation, shows associated with times and concentrations, obviously on 48 h.2..The cell morphology results show that Human recombinant Leptin can induced Hela Cells morphological changes. Control group shows that between the cells are closing and cells are polygon;the others shows that long a fusiform or spindle shape and intercellular broadening; Leptin Ⅲ group the most obvious morphological changes and sparse.3.Western Blot results show that Human recombinant Leptin can activated the PI3K/AKT signaling pathway,up-regulated the expression levels of p-AKT,HIF-1α,VEGF.Control group, the expression levels of p-AKT,HIF-1α,VEGF than the others lower, Leptin Ⅰ group, Leptin Ⅱ group, Leptin Ⅲ group higher. Statistical data analysis suggest that Leptin groups the expression levels of p-AKT,HIF-1α,VEGF than higher the Control group, the differences are statistically significant(p<0.05); Leptin Ⅰ group, Leptin Ⅱ group, Leptin Ⅲ group the expression levels of p-AKT,HIF-1α,VEGF shows a trend of increasing, Leptin Ⅲ group p-AKT,HIF-1α,VEGF expression than Leptin Ⅰ group and Leptin Ⅱ group remarkably higher, the differences are statistically significant(p <0.05),then Leptin Ⅰ group and Leptin Ⅱ group the expression levels of p-AKT,HIF-1α,VEGF have no significant difference(p>0.05)Conclusions: 1.Leptin can induced Hela Cells of Cervical Cancer proliferation, and shows associated with times and concentrations;2. In Hela Cells of Cervical Cancer,Leptin can induced up-regulated the expression levels of HIF-1α,VEGF colud be through up-regulated the expression levels of p-AKT activated the PI3K/AKT signaling pathway.
