Objective:In this study, the renal denervation model of spontaneously hypertensive rats(SHR) were be built.And by comparing the activity of rats RAAS system, heart weight/body weight ratio, and morphological changes in myocardial tissue of rats under the microscope to research the effect on ventricular remodeling in SHR with renal denervation.Methods:1, Grouping and blood pressure measurement. Fifty 12-week-old male spontaneously hypertensive rats (SHR) were randomly divided into the left side dispose group (group L), the right side dispose group (group R), bilateral dispose group (group D), sham operation group (group S) and blank control group (group B), the five ordinary 12-week-old male WKY rats prepared for the negative control group (group F), and the group L, group R and group D are the total combination of dispose group (group M) for statistical analysis. Blood pressure of the rats were measured after completion of grouping.2, rats renal denervation. M group of rats were anesthetized with 10% chloral hydrate by intraperitoneal injection.Then fixed and opened abdominal cavity, separated bilateral renal arteries. According to grouping, stripped left, right or bilateral organization around renal artery under the microscope, and rubbed renal artery with 10% phenol solution.Made flayed tissue with HE staining to confirm containing nerve tissue. S group rats directly reseted abdominal organs and closed after exposed to bilateral renal arteries. After surgery, gave penicillin antibiotics a week, placed under the same conditions with other groups of rats and raised 6 weeks.3, Noninvasive blood pressure measurement in rats.1 weeks,2 weeks,3 weeks,4 weeks, weeks,5 weeks,6 weeks, respectively, measured the blood pressure of rats in each group.4.HW/BW value and the RAAS system activity detected. After 6 weeks the body weight of rats were measured in each group. Then rats were killedandtheir hearts was removed with filter paper, carefully removing the connective tissue around the heart after weighing, calculating heart weight to body weight ratio of HW/BW. Cut the same parts of the small part of the heart tissue in EP tube to detect the myocardial tissue of rats ACE, AngⅠ,AngⅡ, ALD content relative values with western blot method and used SPSS 17.0 statistical analysis software for statistical analysis.4, The thickness of the rat measurement, myocardial fibrosis and tissue morphology observation. The hearts of rats are cut along the largest surface, measuring the thickness of the myocardium at the apex to comparative analysis. The remaining heart tissue made into paraffin sections were stained with HE and masson staining and observed morphological characteristics and fibrosis in rats myocardial tissue under a microscope.Results:1, M rats in the postoperative 1W blood pressure decreased significantly, after the blood pressure gradually increased,6 weeks after the recovery to preoperative level, and the D group blood pressure drop in the level of L group and R group.2. In the M group, the blood pressure gradually increased after the operation, the blood pressure gradually increased, and the blood pressure of the D group was stronger than that of the L group and the R group at 6 weeks after operation.3.HW/BW value, myocardial thickness, cardiac tissue ACE, AngI, AngⅡ, ALD content relative value of group B rats were significantly higher than group M (p <0.05).Under the microscope, the hypertrophy of cardiac muscle cells and the interstitial edema and slight hyperplasia of fibrous tissue were slightly more than M group.4., HW/BW value, myocardial thickness, cardiac tissue ACE, AngⅠ, AngⅡ, ALD content relative value of group S rats were significantly higher than group M (p <0.05). Under the microscope, the hypertrophy of cardiac muscle cells and the interstitial edema and slight hyperplasia of fibrous tissue were slightly more than M group.5, HW/BW value, myocardial thickness, cardiac tissue ACE, AngⅠ, AngⅡ, ALD content relative values of group L or R rats were significantly higher than group D (p <0.05), but there is no significant difference between group L and group R. The degree of hypertrophy and hyperplasia of the fibers were more obvious than that of the D group, but there was no significant difference between the L group and the R group.6, HW/BW value, myocardial thickness, cardiac tissue ACE, AngI, AngⅡ, ALD content relative value of group B rats were significantly higher than in group F (p <0.05). Compared with the M group, the cardiac muscle cells were significantly hypertrophy, the structure was not complete, the myocardial boundaries were not clear, the interstitial was edema and the fibrous tissue was hyperplasia.7, There is no significant difference between group B rats and group S in HW/BW value, myocardial thickness, cardiac tissue ACE, AngI, AngⅡ, ALD relative value content. There was no significant difference in the degree of cardiac hypertrophy and fiber hyperplasia between group B and group S rats.Conclusions:1、RDN can decrease the hypertension in the acute stage, but the blood pressure can be recovered to the preoperative level after a certain period of time2、Renal denervation can obviously relieve hypertension caused by ventricular remodeling in the acute stage.3、The effect of RDN on blood pressure and ventricular remodeling in renal artery was positively correlated with RAAS activity in myocardium4、In the acute stage, the effectiveness of bilateral renal denervation on ventricular remodeling was stronger than unilateral renal denervation.
