Cell death was the end of the life.There were many factors leading to cell death,including harsh environment and the damage factors.There were obvious characteristics in dying cell that could be observed a large number of ROS and damaged seriously of organelles.Cell death has been subdivided of apoptosis,necrosis and pyroptosis.Apoptosis is the process of programmed cell death,that occur dependent of caspase family.Caspase3/7 was the final executant of apoptosis and Z-VAD was the specific inhibitor.Necrosis is a form of traumatic cell death that occurs under a variety of physological and pathological conditions.Necrosis might also be mediated by some programs,RIP3 played key roles in these process.Pyroptosis is a new form of programmed cell death,which is dependent on caspase1 activation and leads to the release of inflammatory cytokines.The molecular mechanisms,morphological and biochemical properties of this form of cell death fire distinct from necrosis or apeptosis.Studies have shown that pyroptosis plays a significant role in a variety of disease.VX-765 could inhibition pyroptosis by inactivating IL-1β proteins.In our study,rLIP could destroy tumor cells morphology and inner structure and induced cell death.Specifically,rLIP induced remarkable cell morphological laterations such as cell blebbing and the plasma membrane was destroyed.In addition,the rLIP induced MCF-7 cells endoplasmic reticulum(ER).Cellular Ca2+ homestasis has been maintained by ER.Many activities of Ca2+ has been associated with ER.The changes of intracellular Ca2+ could induce ER stress and overstress has an determined end of cell death.rLIP also induced MCF-7 cell mitochondria and cytoskeletal structure,further by electron leak of respiratory chain generated reactive oxygen species(ROS).The sigbaling act of ROS is emphasized in cell death.Important inflammatory factors and the protein of cell death were significantly increased by rLIP in tumour cells.In the research,we found that the single closed cell apoptosis,pyroptosis and cell necrosis pathway can not inhibit cell death caused by rLIP.Closing all three pathways of cell death,dying cells were not decreased by rLIP.Further study showed that GPI-APs as the rLIP identifying targeting and revealed that rLIP destruction tumor cells.rLIP could become a potential drug target for these diseases.
