Objective:To explore the immunological regulating effect and mechanism of bone marrow mesenchymal stem cells in mouse allogenic skin transplantation.Method:To isolate and identify mouse BMSCs.Thirty male C57 BC / 6 mice were randomly divided into three groups of ten,namely,syngeneic transplantation group,the control group and the experimental group.In the experimental group,recipients received 2×104/g BMSCs by vein injection in tail two hours after operation while recipients were given 350μL PBS by vein injection in control group and syngeneic transplantation group.Test items include:(1)survival time of skin graft,(2)graft pathology,(3)splenic FOXP3 m RNA expression by Real-Time PCR,(4)Serum levels of IL-2,IFN-γ,TNF-α and IL-4 by ELISA.Result:The survival time of skin graft amounted over twenty days in syngeneic transplantation group.The survival time of skin graft prolonged in control group and the difference was significantly statistical(P <0.05).Graft HE staining: The number of inflammatory cells in the experimental group was significantly less than the control group.The splenic FOXP3 m RNA expression in experimental group was significantly higher(P <0.05)and serum level of IL-4 was significantly increased in the experimental group,whereas serum IL-2,INF-γ and TNF-α decreased significantly in the experimental group(P <0.05).Conclusion:1 We successfully isolated and identified BMSCs;2 We successfully constructed mouse skin transplantation model;3 BMSCs can prolong the survival time of allograft skin,which may be regulate splenic expression of FOXP3 m RNA and secretion of cytokines.
