Objective: To investigate the feasibility of using serum free medium to isolate malignant melanoma stem cells from malignant melanoma cells,and then to study the biological characteristics of malignant melanoma stem cells.Methods: Malignant melanoma tumor cell line A375 cells were inoculated with b FGF and B27,EGF in serum free culture medium to observe the proliferation process,by using serum free culture medium to obtain cell spheroids,the ordinary melanoma cells was injected into the BALB / c mice in vivo compared their induced by tumor and the tumor,immunohistochemistry and detection of tumor stem cell surface marker CD20,CD133 expression,the sphere cells and tumor cells were added to Transwell chamber compared their migration and by immunofluorescence detection of the tumor stem cell surface marker CD20,CD133 expression.Results: The serum-free medium can get spheroids,cellular spheroids induced by tumor and migration ability in ordinary adherent cells and tumor tissue can be detected the expression of CD20,CD133,spheroids CD20,CD133 expression was significantly higher than that of normal adherent cells.Conclusions: Cancer stem cells exist in malignant melanoma and no serum medium sphere cells can be enriched for cancer stem cells,tumor stem cells have a very strong induced by tumor and metastatic ability and sphere cells expression of tumor stem cell marker.
