Aim:Explore a natural physical,quickly and efficiently method to obtain SVF Gel model.Text the biological characterization of the model,which identify the biological characteristics of ADSC,as to provide an access to use SVF / ADSC in an effective way.Methods:1.The establishment of SVF Gel Model: people get fat structure.Preparation of the packet.Straight two-pipe structure connecting the two built-in 10 ml screw-fat 10 ml syringe,that is,different groups to set the volumetric flow rate Q and time t is processed,observe the destruction rate of mature adipocytes.SVF Gel different groups living dead cell assay.SVF Gel different groups in ECM scanning electron microscopy.Flow cytometry analysis of different constituencies ADSC,endothelial cells,blood cells derived from each cell population proportion.2.Identification of SVF Gel Model: SVF SVF Gel in ADSC and Separation and cultured in vitro adipogenic,osteogenic differentiation and orientation detection.Results:1.The same volume flow rate at different injection time effects,different degrees of mature adipocytes destruction of fat tissue.The longer the time,the greater the degree of destruction of mature fat cells,the greater the degree of destruction of the extracellular matrix,wherein at 5min role of the extracellular matrix form stable.Different time volumetric flow rate of 10 m / s under the effect of varying amounts of ADSC was no statistically significant.2.SVF Gel in ADSC isolated and cultured in vitro adipogenic and osteogenic capability.Conclusion:1.Successfully constructed SVF Gel model: volumetric flow rate of 10 m / s,5 min processing structure fat can destroy the greatest degree of mature fat cells,fat droplets get vascular stroma fragments gel.2.SVF Gel in ADSC isolated and cultured in vitro adipogenic and osteogenic capability.
