Objective:Vascular endothelial cell damage and vascular wall structure change caused by hemodynamics may be one of the mechanisms of intracranial aneurysm formation.Vascular endothelial cadherin(VE-cadherin,VE),P120 catenin(P120catenin,P120ctn)and Kaiso play an important role in maintaining the stability of the endothelial cells.The effect of impinging flow on the expression of VE,P120 and Kaiso in human umbilical vein endothelial cells was studied in this study.Methods:The human umbilical vein endothelial cells(HUVEC)were placed in the T type impinging flow cell culture system designed by our research group,which was established to simulate the blood flow at the apex of the vascular bifurcation.Cells were loaded under a steady flow rate of 250 ml / min [equivalent to the Reynolds number(Re)= 125],immunocytochemistry,immunofluorescence and fluorescence quantitative PCR were applied to observe the expression of VE,P120 and Kaiso in HUVEC in different impact time(0 h,4 hours,6 hours and 8 hours).Results:1.The endothelial cells in the vicinity of the impact point maintained fusion,no cell separation,contraction or falling off was observed.2.The expression of VE,kaiso and p120,measured by immunocytochemistry and immunofluorescence technique respectively,decreased with the prolonging of impact time(P < 0.05).3.Fluorescence quantitative PCR technology showed the expression of VE,P120 and Kaiso decreased with the prolonging of impact time.Conclusion:The change of expression of VE,P120 and Kaiso in vascular endothelial cells was earlier than the change of cell morphology.The expression of VE,P120 and Kaiso decreased with the prolonging time of impact.It is indicated that the change of expression of VE,P120 and Kaiso in vascular endothelial cells can be caused by the hemodynamics,which leads to the damage of adherent junction in endothelial cells under impinging flow.
