Objectives: To explore the therapeutic effect of dissipating phlegm and resolving masses method in murine schistosomiasis liver fibrosis and the influence on liver function,fibronectin and TSPO,along with the therapeutic mechanism of treating hepatic fibrosis via the method of dissipating phlegm and resolving masses,so as to provide a theoretical basis for treating liver fibrosis with traditional Chinese medicine from the aspect of “phlegm”.Methods: The models of liver fibrosis induced by schistosoma were established by exposure of mice percutaneously through the tail to cercariae of schistosoma and the dissipating phlegm and resolving masses decoction(the ethanol extract of fritillaria and rhaponticum uniflorum)were produced into three kinds of low,medium and high doses(50mg/kg,100mg/kg,150mg/kg).The mice were randomly divided into normal control group(group A),model control group(group B),low dose dissipating phlegm and resolving masses decoction group(group C),middle dose dissipating phlegm and resolving masses decoction group(group D)and high dose dissipating phlegm and resolving masses decoction group(group E).From the beginning of the sixth week,we begin to treat those mice in group C,group D and group E with low,middle and high dose dissipating phlegm and resolving masses decoction and other mice in group A and group B with distilled water.The period of Treatment were eight weeks.After the end of treatment,we collected serum samples for one to two milliliter and killed the mice by Cervical dislocation method.Then we got the samples of liver tissue by the dissection of mice.The serum transaminase were detected by automatic biochemical analyzer and histological sections of liver were evaluated by hematoxylin and eosin stains.The content of fibronectin were assayed by Real-time Polymerase Chain Reaction and the expression of TSPO were examined by Western Blot.Results:1.In the sixth week,we observed the histological sections of liver which were stained by hematoxylin and eosin and found the formation of egg granulomas and the regeneration of nodules.we also found fibrous connective tissue hyperplasia and inflammatory response in the portal of the liver.These were strongly proved that the models of liver fibrosis induced by schistosoma were established successfully.2.After the end of the treatment,compared with the normal control group,the level of serum ALT and AST in the model control group were significantly increased(P<0.01).Compared with the model group,the level of serum ALT and AST in group C,group D and group E were decreased obviously(P<0.05).Comparison among the three groups C,D and E,there were a statistically significant difference between them(P<0.05).Among them,the level of serum ALT and AST in group E were most noticeably decreased,group D were followed by.3.After the end of the treatment,compared with the model control group,the areas of egg granuloma in group A,group B and group C were decreased obviously(P<0.05).Comparison among the three groups C,D and E,there were a statistically significant difference between them(P <0.05).Among them,the areas of egg granuloma in group E were most noticeably reduced,group D were followed by.4.After the end of the treatment,compared with the normal control group,the level of fibronectin in liver tissues in the model control group were significantly increased(P<0.01).Compared with the model group,the level of fibronectin in liver tissues in group C,group D and group E were decreased obviously(P<0.05).Among them,the level of fibronectin in liver tissues in group E were most noticeably decreased,group D were followed by.5.After the end of the treatment,compared with the normal control group,the expression level of TSPO in liver tissues in the model control group were significantly increased(P < 0.01).Compared with the model group,the expression level of TSPO in liver tissues in group C,group D and group E were decreased obviously(P<0.05).Conclusions:1.Under normal circumstances,exposure of mice percutaneously through the tail to cercariae of schistosoma for about six weeks can successfully set up the model of hepatic fibrosis and in generally it was better to infect about fifteen cercariaes.2.The method of dissipating phlegm and resolving masses can effecti-vely treat on murine schistosomiasis liver fibrosis.3.The method of dissipating phlegm and resolving masses have obviously inhibitory effect in the liver fibrosis induced by schistosoma infection.The mechanisms maybe related to protect the liver function,inhibit the inflamma-tion of the liver,reduce the expression of FNmRNA and TSPO.
