| Objective1.To investigate the expression of transferrin receptor(TfR), ferritin(FN) and autophagy-related genes Beclin1 and LC3 in mRNA level in the bone marrow mononuclear cells of children with acute leukemia(AL).2.To preliminary study the relationship between autophagy and iron metabolism in children with AL, and to provide a new strategy for AL targeted therapy in children. MethodsTo collect 46 bone marrow specimens from children with acute leukemia from one hospital pediatrics hematology ward in Hennan from October 2014 to October 2015.Patients were divied into 2 groups, 30 cases of ALL, 16 case of AML.A group of 16 healthy children was used as a control. Bone marrow mononuclear cells(BMMNCs) were separated by lydroxypropylmethyl cellulose.RT-PCR was used to detect the expression levels of TfRmRNA, FNmRNA, Beclin1 mRNA and LC3 mRNA and TfRmRNA in every group. Two independent sample t test was used to analyze the expression difference, and Pearson correlation analysis was performed to evaluate the correlations coefficient between them,α=0.05. Results 1.The general situation of the inclusion of the objectThe difference about gender(χ2=1.033,P=0.309),age(t=0.269,P=0.798) was not statistically significant in ALL group and the control group. The difference about gender(χ2=0.139,P=0.710),age(t=0.241,P=0.811) was not statistically significant in AML group and the control group. 2.Expression levels of autophagy-related genes Beclin1 mRNA,LC3 mRNAThe expression levels of Beclin1 mRNA in ALL group,AML group and the healthy control group were(0.945±0.071)(0.632±0.068)(0.759±0.072);the expression levels of LC3 mRNA were(0.416±0.068)(0.171±0.045)(0.226±0.065). The expression of Beclin1 mRNA and LC3 mRNA in ALL group were higher than the control group(t=8.422,P<0.001)(t=9.128,P<0.001),but in AML group were lower than the control group(t=-5.178,P<0.001)(t=-2.781,P=0.009),The differences all were statistically significant. 3.Expression levels of TfRmRNA and FNmRNAThe expression levels of TfRmRNA in ALLgroup,AML group and the healthy control group were(1.835±0.072)(1.968±0.115)(0.991±0.096);the expression levels of FNmRNA were(0.323±0.062)(0.426±0.055)(0.217±0.039).The expression of TfR mRNA and FNmRNA in ALL group were higher than the control group(t=33.637,P<0.001)(t=6.374,P<0.001),and in AML group were higher than the control group(t=26.178,P<0.001)(t=12.407,P<0.001),The differences all were statistically significant. 4.The relationship between TfRmRNA,FNmRNA and autophagy-related genes Beclin1 mRNA and LC3 mRNAIn ALL group,AML group and the healthy control group, TfR levels inversely correlated with Beclin1(r=-0.899,P<0.001)(r=-0.876,P<0.001)(r=-0.895,P < 0.001); TfR levels inversely correlated with LC3( r=-0.925, P < 0.001)(r=-0.923,P<0.001)(r=-0.938,P<0.001); FN levels positively correlated with Beclin1(r=0.969,P<0.001)(r=0.906,P<0.001)(r=0.879,P<0.001); FN levels positively correlated with LC3(r=0.947,P<0.001)(r=0.894,P<0.001)(r=0.924,P<0.001). Conclusions1.In both acute leukemia patients and control group,TfR,FN and autophagyrelated genes Beclin1 and LC3 are expressed, But in acute leukemia cells, FN, TfR expressions are significantly increased;autophagy related genes Beclin1 and LC3 expressions are increased in ALL, while in the AML,the expressions are decreased suggesting that the abnormity of autophagy and iron metabolism may play an important role in the pathogenesis of acute leukemia.2.In both acute leukemia patients and control group,TfR levels inversely correlate with autophagy-related genes Beclin1 and LC3,but FN levels positively correlate with autophagy-related genes Beclin1 and LC3,which shows that there is some correlation between autophagy and iron metabolism. |
