| Background:Liver cancer is to point to by liver cells or intrahepatic bile duct epithelial cells of cancer,it is one of the most ordinary mailignancy in china,it comes off second best in all mailignancies.The world each year,25000 people died of liver cancer.40% of them is in china.Because of with rapid growth of liver cancer,invasion obilty,high postoperative recurrence and resistance to chemotheropy drugs.The 5-year survival rate of only 15%,advanced liver cancer the median surial only six months in the current strategies.So how to be more effective in the treatment of liver cancer and improve patients survival and quality of life has become very urgent.Cell apoptosis is triggered by factors in vivo and in vitro cell death process,in order to keep the body and since the stability of the organization.Development and metastasis of tunor is closely related to the lack of cell apoptosis become an important effective way of treatment of liver cancer. Objective:Explore flavonoids of artemisia argy to inhibit the growth of liver cancer smmc-7721 cells and induce apoptosis;Research flavonoids of artemisia argy induced liver cancer smmc-7721 cells apoptosis and caspase-3、p21、bcl-2 apoptosis protein,and the relationship between the three. Methods:Developing liver cancer smmc-7721 cells,flavonoids of artemisia argy is used in liver cancer smmc-7721 cells for 48 hours,cck-8 method to detect cell inhibition rate,flow cytometry instrument measuring each cell apootosis rate,RT-PCR method detect the expression of caspase-3、p21、bcl-2 m RNA. Results:Flavonoids of artemisia argy inhibit liver cancer smmc-7721 cells proliferation and promote apoptosis.Cell inhibition and apoptosis rate,in time,the concentration dependence.Flavonoids of artemisia argy is used in liver cancer smmc-7721 cells for 48 hours,the mRNA of caspase-3、p21 express to rise,,the mRNA of bcl-2 express to descend., Conclusions:Flavonoids of artemisia argy inhibit liver cancer smmc-7721 cells proliferation and promote apoptosis,and through signal way of about caspase-3、p21、bcl-2 induced its aopotosis. |
