Sophora flavescens Ait, is highly respected in pharmaceutical industry at both domestic and abroad. Early studies on chemical compositions of Sophora flavescens. concentrated on alkaloids. Since the beginning of the 1970s, studies on chemical compositions and pharma-cological effects of flavonoids in Sophora flavescens Ait. have gradually increased and tremendous results have been received. The results showed that flavonoids in Sophora flavescens Ait. had a wide range of physiological effects and medicinal value. In recent years, many researchers are trying to use different methods to extract and separate total flavonoids which have medicinal value from Sophora flavescens Ait.. Whereas traditional methods of extraction and separation of total flavonoids have many shortcomings, for example, they have many procedures, the extraction rate is low, they waste a lot of organic solvent in extraction process, ect. This experiment aimed to explore the chemical property differences in total flavonoids content based on numerous extraction and separation methods. It used alkali extraction and acid precipitation method to extract total flavonoids and new technology to purify ethanol. The experiment was successful and achieved the goal of high yield rate, high purity, low energy consumption, low cost, low environmental pollution. With the development of the research on flavonoids in Sophora flavescens Ait., researchers found that flavonoids in Sophora flavescens Ait. had a wide range of pharmacological activity. But there were few studies on its antimicrobial evaluation and determination methods. This experiment optimized the extraction process of total flavonoids in Sophora flavescens Ait. and studyed the impact of total flavonoids on different cultures. It was important for discovering new flavonoids drugs or new guided compounds. It can also provide theoretical basis for the further development and utilization medicinal value of Sophora flavescens, and provide experimental evidence for the bacteriostatic effects of total flavonoids in Sophora flavescens Ait.First, optimize the extraction conditions of total flavonoids in Sophora flavescens Ait.:the total flavonoids content and extraction rate were taken as indexes to analyze the alkali pH value, acid pH value, ratio of solid to liquid, extraction time of percolation extraction method, respectively. We analyzed investigation results by orthogonal test, purified total flavonoids and determined the optimum condition. The optimized percolation extraction process is:fully infiltrate Sophora flavescens Ait. with 0.2% aqueous hydrochloric acid solution for 2 hours, make sure the ratio of solid to liquid is 1:15 using percolation method, percolate for 24 hours to remove alkaloids in Sophora flavescens Ait. and adjust the pH value of the wet ingredients, then use sodium hydroxide solution to percolate, ratio of solid to liquid is 1:20, percolate for 36 hours, make sure the pH value of percolation solution is 10 and pH value of acid precipitation is 4. The optimized purification condition of total flavonoids in Sophora flavescens Ait. is:mix crude solution of total flavonoids with 90% ethanol solution, reflux extract total flavonoids by 90℃ water bath four times, then we can get pure total flavonoids. Compared with corresponding alkaloids in herbs, the average extraction rate of total flavonoids in 10 small-scale tests was 80.46% and the average purity of total flavonoids 90.32%.We identified every physical and chemical indicators of total flavonoids in Sophora flavescens Ait. measured by HPLC method, This process extracts the ingredients prove indeed Total flavonoids of Sophora flavescens. HPLC chromatograms of shows, process maps is better, The main ingredient kurarinone is approximately 43.10% of total flavonoids. They all showed that the above-mentioned extraction and separation process of alkaloids in Sophora flavescens Ait. was successful. But it has not been reported.Then, we studied the antibacterial properties against six kinds of bacteria of total flavonoids in Sophora flavescens Ait. and measured the minimum inhibitory concentration.Experiments showed that the inhibitory concentrations of total flavonoids in Sophora flavescens Ait. against Staphylococcus aureus, Staphylococcus, Bacillus subtilis, E. coli, lemon Staphylococcus aureus, Pseudomonas aeruginosa and other bacteria (mg/ml) were: 2.34,2.56,2.28,4.67,4.35,4.31.Total flavonoids of Sophora flavescens are salinogenic under alkaline conditions and are water-insoluble under acidic conditions. The alkaline solution can help to extract alkaloids and alkaloids in Sophora flavescens Ait. can dissolve under acidic conditions, so we can use acid solution percolation method to remove alkaloids in Sophora flavescens Ait, then use alkali extraction and acid precipitation method to percolate and extract total flavonoids in Sophora flavescens Ait. Through which we can maximize the extraction rate, reduce impurities and improve the utilization of medicines. Total flavonoids in Sophora flavescens Ait has bacteriostatic and bactericidal effects on the growth of above-mentioned bacteria, and they are more effective against gram-positive bacteria than Gram-negative bacteria. The experiment proved that taking appropriate extraction and separation methods on the basis of different physicochemical properties caused by different chemical structurals is feasible. This method has many advantages such as simple process, low cost, high purity of the total flavonoids, low environmental pollution, and it is suitable for industrial production.
