Study On The Qingre Jieji Granule Volatile Oil Extraction Clathration Craft And Quality Control

1 Objective1. The optimal extraction conditions of volatile oil from Chinese medicinal herbs were selected and the volatile oils were identified qualitatively. 2. Process parameters for the inclusion of volatile oil beta-cyclodextrin inclusion complex and qualitative identification of volatile oil inclusion compound. 3. Content of volatile oil in the extract of volatile oil, volatile oil inclusion compounds as well as Qingre jieji particles were determined, and stability of three batch of Qingre jieji particle content were inspected.2 Method1.With the amount of volatile oil as the standard, it was obtained that the best technological conditions for the extraction of volatile oil from Bupleurum,notopterygium, ginger, angelica, and studies the contents of volatile oil in the medicinal materials through using single factor test. 2.The effects of the inclusion temperature, inclusion time, the ratio of cyclodextrin and volatile oil on the inclusion results were investigated by orthogonal test. The optimal inclusion conditions of volatile oil beta cyclodextrin inclusion complexes were screened and the inclusion test was performed. 3. Volatile oil content of medicine herbs extracts, cyclodextrin inclusion compound of volatile oil and Qingre jieji particles were qualitative identificated by thin layer chromatography. 4. Chemical constituents of the volatile oil from Chinese medicinal herbs were separated and identificated by gas chromatography and mass spectrometry while the index components were selected;5.Content of volatile oil of medicine herbs extracts, cyclodextrin inclusion compounds of volatile oil, Qingre jieji particles were determinated by gas chromatograph, and then the realted methodology research was successfully compleletd. 6.The stability of volatile oil in the three batch of samples were tested for long-term retention test and accelerated test.3 Result1. The best extraction technology of the four herbs included that, i)notopterygium : 8 times water continuous reflux extraction 6h; ii) Ginger : 8 times water, continuous reflux extraction 7h; iii) Angelica: 6 times water, continuous reflux extraction 5h; iv) Bupleurum: 6 times water, continuous reflux extraction 6h. The optimal extraction condition of volatile oil from Chinese medicinal herb was add 8times of water continuous reflux extraction 6h. 2. Notopterygium root, dahurian angelica root, ginger, North Bupleurum four herbs of the volatile oil content limits were respectively 1.55%(m L / g), 0.21%(m L / g), 1.65%(m L / g), and 1.12%(m L / g).3. The optimal extraction conditions of volatile oil from Chinese medicinal herbs were that 8 times of water, 6 hours of continuous reflux extraction. 4. Inclusion conditions for the inclusion compound of volatile oil were volatile oil and beta cyclodextrin ratio1:8, add 1 times ethanol soluble volatile oil, under the 50 ℃ temperatures to clathration 1 hour. 5. TLC results showed that beta-pinene and cajeputol could be qualitatively and quantitatively identificated from volatile oil of medicine herbs extracts. 6.The results showed that the GC method established in this study was accurate, reliable and feasible for the determination of volatile oil content.7.Accelerated test and long-term retention test results showed that main components content within the volatile oil decrease compared to 0 month.4 Conclusion This experiment research outcomes for Qingre jieji granule of volatile oil content quality standards provides certain theoretical basis. The experimental results showed that the preparaed process is reasonable, feasible, and reliable. And detection analysis method is convenient, accurate, and reliable.
