| Objective Observation of Electroacupuncture at Jing Points on vascular vascular dementia(VD)model of learning and memory in rats hippocampal CA1 region of p38mitogen-activated protein kinase(p38MAPK) expression and apoptosis Explore the possible mechanism electric about Jing Points on the VD in order to provide some experimental and theoretical basis for the clinical treatment of acupuncture points VD.Methods1. Choose clean adult male Wistar rats 70, weighing(275 ± 25) g, after adaptive feeding one week, then use particularly sensitive screening test jumping too slow and large mouse,so that can reduce animal avoidance response differences caused by differences,60 retention, Computer random number generation method sham group 8,the remaining rats using a modified four-vessel occlusion(4-VO) to prepared of VD model, four days after the replication model,using step down test and neurobehavioral to determined whether the model are successfully copied or not.the computer randomly divided into acupuncture group and western medicine group,model control group 3 groups, each 8. After 10 days, EA group were given acupuncture treatment, day 1, 10 days for a course of treatment, a total of three courses, western medicine group were given piracetam oral treatment, twice daily treatment with acupuncture group parallel, model control group and the sham group are regular feeding,and donot with special treatment.2.Experimental and neurobehavioral scores measured and drawn using immunohistochemical method to detect the expression of hippocampal CA1 region p38 MAPK, Tunel assay rats hippocampal CA1 region apoptosis. Using SPSS17.0statistical analysis of experimental data.ResultsAfter the model copying 4d, using a step-down test to detect the incubation period,the number of errors and detects neurobehavioral scores. Results: The model group and the sham group replication latency reduced(P<0.01) as compared to the number of errors increased(P<0.05), neurobehavioral scores are increased(P<0.05), The difference was statistically significant; model control group Western medicine group reduced latency compared(P<0.05), the difference was statistically significant, the number of errors increased(P>0.05), neurobehavioral scores(P>0.05), the difference was not statistically significant, electro-acupuncture group and western medicine group phase than the latency(P>0.05), reduce the number of errors(P>0.05), neurobehavioral scores(P>0.05), the difference was not statistically significant. The results showed that EA group could improve the learning and memory in rats with VD and neurobehavioral science scores,and EA group was superior to western medicine group in improving learning and memory ability.After treatment, using immunohistochemical method to detect the expression of p38 MAPK in hippocampal CA1 region. Comparing with the model group, Sham group mean gray values are increased(P<0.01), positive cells are reduced; compared with Western medicine group, reducing the average gray value model control group(P <0.01), the difference was statistically meaning reduced compared with the average gray value EA group(P<0.01), an increase in positive cells; Western medicine group and EA group, the gray level(P>0.05), the difference was not statistically significant; EA group compared with the sham group, the mean gray values are increased(P<0.05), the results showed that the positive expression in hippocampal CA1 region of p38 MAPK EA group can be reduced.The results show that using electric on Jing Points can inhibit the degree of hippocampal neuronal apoptosis.Tunel assay apoptosis in hippocampal CA1 region, as compared to the apoptosis index decreased sham group and model control group(P<0.01), the difference was statistically significant, the degree of apoptosis was significantly reduced; electro-acupuncture group and model control compared to the group, to a lesser extent apoptosis(P<0.01), the difference was statistically significant; Western medicine group and the model control group, reduce the degree of apoptosis(P<0.01), the difference was statistically significant; EA group compared with sham,apoptosis index increased(P<0.01), the difference was statistically significant,western medicine group compared with the sham group, apoptotic index increased(P<0.01), the difference was statistically significant. Western medicine group and EA group, apoptotic index increased(P<0.01), the difference was statistically significant, the results indicate that acupuncture can reduce the well hole in hippocampal CA1 AREA index, reduce neuronal apoptosis, and effective than Western medicine group.Conclusion1. Electrical Jing Points can improve learning and memory abilities of VD rats, and can reduce VD neurological deficit scores.2. acupuncture points can inhibit the expression of p38 MAPK well.It was suggest that acupuncture may well Points VD treatment by inhibiting p38 MAPK signal path, and reducing neuronal apoptosis.3. Using electric on Jing Points can reduce apoptosis in rat hippocampal CA1 region VD, reduce secondary damage and promote neuronal repair. |
