Sophora tonkinensis Gapnep. and Cimicifuge racemosa(L.)Nutt.is an important traditonal chinese herbal medicine, which have strong anti-tumour. Sophora tonkinensis Gapnep. contains flavonoids have good anti-ulcer activity and Cimicifuge racemosa(L.)Nutt. contains saponins have better mitigation effect of menopause. To separate the flavonoids from Sophora tonkinensis Gapnep. and the saponins from Cimicifuge racemosa(L.)Nutt. in order to investigate pharmacological activity and structure-activity relationship.The flavonoids of Sophora tonkinensis Gapnep. were isolated by using silica gel column chromatography, reversed-phase column, Sephadex LH-20, high performance liquid chromatography, crystallization and re-crystallization, And their structures were elucidated on the basis of NMR, MS, and UV.The dichloromethane portion of Sophora tonkinensis Gapnep. were isolated and repeatedly purified by various chromatographic techniques and recrystallization above mentioned. Eight flavonoids were isolated and identified as sophoranone(1, 1.300 g), lespeflorin A3(2, 0.810 g),L-Maackiain(3, 0.325 g), glabrol(4, 0.056 g), sophoradin(7, 0.051 g),sophoranochromene(8, 0.750 g), Lupinifolin(9, 0.015 g), 5-Hydroxysophoranone(10, 0.18 g), and Three non-flavonoids were isolated and identified as β-sitosterol(5, 0.098 g), lupeol(6, 0.043 g) and soyasapogenol B(11, 0.010 g). Compound 2 is isolated from this plant for the first time. Compounds 10 are firstly isolated from this genus.The Ethyl acetate portion of Cimicifuge racemosa(L.)Nutt. Were isolatted and repeatedly purified by column chromatography and recrystallization, and their structures were identified by physicocheical properties and spectral data. Two saponins were isolated and identified as actein(0.208 g), 27-deoxyactein(0.017 g). The purity of actein and 27-deoxyactein was98.2%, 98.3%. The total yield of actein was 0.020%, including 0.001% 27-deoxyactein.
