Armillarialuteo-virens, belonging to Tricholomataceae, Armillaria, was also known as yellow mushroom, golden mushroom. Armillarialuteo-virens is a kind of valuable wild edible fungus which is rich of nutritious components and high medical value. At present, the material based research of Armillarialuteo-virens mainly concentrated on volatile composition, polysaccharide and exogenous lectins. However the monomer compound of the fruiting body of Armillaria luteo-virens was still in blank.In this study, we firstly developed a two-dimensional(2-D) preparative liquid chromatography method to purify constituents from the water extract of Armillaria luteo-virens. Using separation mode such as RPLC/ HILIC, RPLC/ NPLC and RPLC/ RPLC, six compounds were isolated. The structures of constituents were identified as pyroglutamic acid(1),uridine(2), 2’-deoxyuridine(3), guanosine(4), inosine(5), adenosine(6) on the basis of physicochemical and spectroscopic analyses.Secondly, we put the precipitation of water extraction soaked with acetone, then we got the acetone extrat of Armillarialuteo-virens. With separation mode NPLC/ HILIC, three sterol compounds which had inhibiting activity on proliferation of HepG2 liver cell were isolated and purified. Their structures were identified as ergoterol(7)、3β-hydroxy-(22E, 24R)-ergosta-5,8,22-trien-7-one(8)、(3α, 5α),(8β, 11β)-diepidioxy-ergost-22E-en-12-one(9) on the basis of physicochemical and spectroscopic analyses. All the compounds were purified from fruit bodies of Armillaria luteo-virens for the first time.In addition, antioxidant activity screening technique and Real-time dynamic cellular analysis( RTCA) were used for detecting the bioactivity of water extract and acetone extract from Armillarialuteo-virens. The result showed that STW_Fr.1~Fr.14 and BT_Fr.7~Fr.11, BT_Fr.13 and BT_Fr.14 had significant antioxidant activity, BT_Fr.7-4, BT_Fr.8-1 and BT_Fr.8-4 have significant inhibition for the proliferation of HepG2 cell. This study can provide scientific reference for pharmacological research and further utilization of wild Armillarialuteo-virens resources.
