Objective: Stanniocalcin1 1(STC1)is found that STC1 is highly expressed in many human cancers and is closely related to tumor apoptosis and invasion.The purpose of this study was to analyze the expression of STC1 protein in papillary thyroid carcinoma,and to explore the relationship between the expression of protein and the clinical pathological features,and to provide a new way for the diagnosis and treatment of thyroid tumors.Method: We collected thyroid carcinoma papillary(PTC)in 19 cases,65 cases of nodular goiter(goiter nodular,NG)and 19 cases of normal thyroid tissues(tissue normal,NT).Proper paraffin block was collected to perform routine HE staining and immunohistochemical staining.We sum up the clinical and pathological characteristics of thyroid papillary carcinoma,and the expression of STC1 protein was detected by Western blot method.SPSS.17.0 for Windows software was used for the result analyses.Values of P<0.05 were considered significant.Result:1 Immunohistochemistry showed that the positive products of STC1 were mainly localized in the cytoplasm and occasionally in the cytoplasm of follicular cells.It was different from staining degree,and was granular brown.The STC1 positive rate in papillary thyroid cancer group was 61.5%;which was higher than that of nodular goiter(26.3%,P<0.05/3)and normal thyroid tissue(15.8%,P<0.01).2 thyroid papillary carcinoma patients with clinical pathological features: papillary thyroid carcinoma lymph node metastasis STC1 positive rate was 80.0%,significantly higher than the thyroid cancer without lymph node metastasis(40.0%),the difference was statistically significant(χ~2=10.920,P< 0.01).The positive rate of STC1 was 45.0%,significantly lower than that of the patients with stage(88%,P<0.01),the expression of STC1 was related with calcification,while it was not associated with age,gender and tumor size in patients with papillary thyroid carcinoma.3 Expression of STC1 protein in three groups of thyroid tissues: the expression of STC1 protein can be detected in 103 cases,and the expression of STC1 protein in papillary thyroid carcinoma was also significantly higher than that in nodular goiter and normal thyroid tissues.The relative expression levels of STC1(STC1/β-actin)were statistically analyzed.The relative expression of STC1 protein in papillary thyroid carcinoma(0.647±0.075)was significantly higher than that of nodular goiter(0.284±0.036,P<0.01)and normal thyroid tissue(0.255±0.057,P<0.01).The difference was not statistically significant(P>0.05)in the nodular goiter group and the normal group.Conclusion:1 The expression rate of STC1 in papillary thyroid carcinoma was significantly higher than that in nodular goiter group and normal group,which was helpful to the diagnosis of papillary thyroid carcinoma.2 The expression of STC1 was correlated with calcification,the clinical stage and lymph node metastasis of papillary thyroid carcinoma,which indicated that STC1 can be used as an effective predicted index for the diagnosis of thyroid papillary carcinoma.3 The positive expression of STC1 in is expected to become a sensitive biomarker of papillary thyroid carcinoma. |
