Objective we use invigorating kidney and marrow joint MSCs to discuss whether they affect expression of P16,P21.The traditional Chinese medicine combined with stem cell research, provide experimental basis for clinical treatment of age-related diseases.Methods 1.Establishment of aging model.SD rats daily subcutaneous injection of D-galactose,4months,And to identify whether the preparation of aging model success.2.The whole bone marrow adherent centrifugation cultivation of MSCs, MSCs was identified by morphologic characteristics, surface markers detected with flow cytometry.3.Observation the groups general situation, the water maze experiment testing rats learning and memory ability, extraction of rat hippocampus using western blot to detect the expression of P16 and P21.Results 1.Adherent cultured MSCs form a spindle, stem cell colony formation, flow cytometry instrument to detect cell surface antigen CD44, CD71 surface expression of positive.2.The treatment groups reach platform number are more than the model group, and the incubation period significantly shorten, there are significant difference.3.Western blot results show that the treatment groups of P16, P21 protein expression is lower than the D-galactose model group, including invigorating kidney+ bone marrow mesenchymal stem cells in the lowest group of P16, P21 protein expression.Conclusion Invigorating Kidney of TCM and bone marrow mesenchymal stem cell transplantation can improve aging rats learning and memory ability, cut the expression of P16 and P21 may be one of the mechanism of reverse aging.
